Neophytadiene (NPT) is a diterpene found in the methanolic extracts of Crataeva nurvala and Blumea lacera, plants reported with anxiolytic-like activity, sedative properties, antidepressant-like actions; however, contribution neophytadiene to these effects unknown. Marine algae have excellent bioresource properties with potential nutritional and bioactive therapeutic benefits, but studies regarding Caulerpa lentillifera are limited. This study aims to explore the metabolites profile antioxidant, anticancer, anti-obesity, in vitro cytotoxicity of fractionated ethanolic extract C. using two maceration soxhlet extraction methods. Dried simplicia was mashed extracted ethanol solvent, concentrated evaporated, then sequentially partitioned equal volumes ethyl acetate n-Hexane. Six samples were used this study, consisting ME (Maceration-Ethanol), MEA (Maceration-Ethyl Acetate), MH (Maceration-n-Hexane), SE (Soxhletation-Ethanol), SEA (Soxhletation-Ethyl SH (Soxhletation-n-Hexane). Non-targeted metabolomic profiling determined LC-HRMS, while anticancer DPPH ABTS, lipase inhibition, MTT assay, respectively. demonstrates that has several functional metabolites, antioxidant capacity (EC50 is very close EC50 Trolox), as well anti-obesity < orlistat, an inhibitor lipid hydrolyzing enzymes), which useful precursors for new approaches improving obesity-related diseases. Obesity is a multifactorial disease characterized by an excessive accumulation of fat, which in turn poses significant risk to health. Bioactive compounds obtained from macroalgae have demonstrated their efficacy combating obesity various animal models. The green Caulerpa lentillifera (CL) contains numerous active constituents. Hence, the present study, we aimed elucidate beneficial anti-obesity effects extracts derived C. using Caenorhabditis elegans model. ethanol (CLET) and ethyl acetate (CLEA) caused decrease fat consumption, reaching up approximately 50–60%. Triglyceride levels 50 mM glucose-fed worms were significantly reduced 200%. GFP-labeled dhs-3, marker for lipid droplets, exhibited reduction its level 30%. Furthermore, intracellular ROS displayed 18.26 23.91% high-glucose-fed treated with CL extracts, while lifespan remained unchanged. Additionally, mRNA expression genes associated lipogenesis, such as sbp-1, showed down-regulation following treatment extracts. This finding was supported (at 16.22–18.29%) sbp-1 gene expression.
