Cancer poses a significant challenge to global public health, seriously threatening human health and life. Although various therapeutic strategies, such as chemotherapy (CT), radiotherapy, phototherapy, starvation therapy, are applied cancer treatment, their limited effect, severe side effects, unsatisfactory drug release behavior need be carefully considered. Thus, there is an urgent develop efficient delivery strategies for improving treatment efficacy realizing on-demand delivery. Notably, pillararenes, emerging class of supramolecular macrocycles, possess unique properties highly tunable structures, superior host-guest chemistry, facile modification, good biocompatibility, which widely used in therapy achieve controllable reduce the toxic effects on normal tissues under internal/external stimuli conditions. This review summarizes recent advance stimuli-responsive systems (SDSs) based pillararenes tumor from perspectives different assembly methods hybrid materials, including molecular-scale SDSs, nano self-assembly systems, nanohybrid SDSs. Moreover, prospects critical challenges SDSs also discussed.

Abstract The combination of macrocyclic chemistry with co-crystal engineering has promoted the development materials vapochromic behaviors in supramolecular science. Herein, we develop a macrocycle based on hybrid[4]arene and 1,2,4,5-tetracyanobenzene that is able to construct materials. Interestingly, can separate benzene/cyclohexane equal-volume mixture allow be removed toluene/ pyridine purities reaching 100%. Selective generation of sufficient pyroptosis inducers at the tumor site without external stimulation holds immense significance for a longer duration immunotherapy. Here, we report cascade-amplified inducer CSCCPT/SNAP that utilizes reactive nitrogen species (RNS), self-supplied from diffusion-controlled reaction between oxygen (ROS) and nitric oxide (NO) to potentiate immunotherapy, while both endogenous mitochondrial ROS stimulated by released camptothecin NO initiate pyroptosis. In recent years, lipid nanoparticles (LNPs) have emerged as a revolutionary technology for vaccine delivery. LNPs serve an integral component of mRNA vaccines by protecting and transporting the payload into host cells. Despite their prominence in vaccines, there remains notable gap our understanding potential application delivery DNA vaccines. this study, we sought to investigate suitability leading LNP formulations plasmid (pDNA). addition, aimed explore key differences properties popular when delivering either or DNA. To address these questions, compared three encapsulating mRNA- pDNA-encoding firefly luciferase based on potency, expression kinetics, biodistribution, immunogenicity. Following intramuscular injection mice, determined that RNA-LNPs formulated with SM-102 ALC-0315 lipids were most potent (all p-values < 0.01) immunogenic 0.05), while DNA-LNPs demonstrated longest duration signal.
