Human osteogenic differentiation is a complex and well-orchestrated process which involves plethora of molecular players cellular processes.A growing number studies have underlined that circular RNAs (circRNAs) play an important regulatory role during human differentiation.CircRNAs are single-stranded, covalently closed non-coding RNA molecules acquiring increased attention as epigenetic regulators gene expression.Given their intrinsic high conformational stability, abundance, specificity, circRNAs can undertake various biological activities in order to regulate multiple processes, including differentiation. The most recent evidence indicates control osteogenesis by preventing the inhibitory activity miRNAs on downstream target genes, using competitive endogenous mechanism. The aim this review draw currently known mechanisms differentiation.Specifically, we provide understanding advances research conducted mesenchymal stem cell types importance regulating osteogenesis.A comprehensive underlying circRNA will improve knowledge processes bone growth, resulting potential development novel preclinical clinical discovery diagnostic therapeutic tools for disorders.

Bladder cancer (BC) is a common and serious type of that ranks among the top ten most prevalent malignancies worldwide. Due to high occurrence rate BC, aggressive nature cells, their resistance medication, managing this disease has become growing challenge in clinical care. Long noncoding RNAs (lncRNAs) are group RNA transcripts do not code for proteins more than 200 nucleotides length. They play significant role controlling cellular pathways molecular interactions during onset, development progression different types cancers. Recent advancements high-throughput gene sequencing technology have led identification various differentially expressed lncRNAs which indicate abnormal expression. In review, we summarize these been found impact several functions related including proliferation, cell growth, migration, metastasis, apoptosis, epithelial-mesenchymal transition, chemo- radio-resistance. The goal this study was clarify essential functions regulating OC progression its underlying mechanism.Relative levels normal tissues were identified by quantitative real-time polymerase chain reaction (qRT-PCR). regulatory effects on proliferation apoptosis SK-OV-3 cells evaluated. Moreover, bioinformatics tools used predict novel target [microRNA (miRNA)] GAS5. To explore key GAS5/miRNA-23a/WT1 loop mediating progression, we performed functional experiments dual-luciferase reporter (DLR) gene assessment.Downregulation found OC, which positively correlated with poor prognosis. In addition, lower expression accelerated migration demonstrated percentage vitro experiments. It acts as molecular sponge for miR-23a cells. Additionally, WT1 detected cells, through sponging miR-23a, regulated expression. Abstract Background The limited therapies available for treating Merkel cell carcinoma (MCC), a highly aggressive skin neoplasm, still pose clinical challenges, and novel treatments are required. Targeting retinoid signalling with retinoids, such as all‐ trans retinoic acid (ATRA), is promising clinically useful antitumor approach. ATRA drives tumour differentiation by modulating signalling, leading to anti‐proliferative pro‐apoptotic effects. Although dysregulated in MCC, activity this unknown. This study aimed evaluate the impact of on pathological phenotype MCC cells. Methods effect was tested various polyomavirus‐positive polyomavirus‐negative lines terms proliferation, viability, migration clonogenic abilities. In addition, cycle, apoptosis/cell death gene signature were evaluated upon treatments. Results efficiently impaired proliferation viability A strong reducing clonogenicity determined ATRA‐treated Moreover, resulted strongly effective arresting cycle inducing all Enrichment analyses indicated that cells promote pathways, which led pro‐apoptotic/cell Conclusions These results underline potential retinoid‐based therapy management might open way experimental approaches other retinoids and/or combinatorial
