The study of biological systems in a holistic manner (systems biology) is increasingly being viewed as necessity to provide qualitative and quantitative descriptions the emergent properties complete system. Systems biology performs studies focussed on complex interactions system components; emphasising whole rather than individual parts. Many perturbations mammalian (diet, disease, drugs) are multi-factorial small parts insufficient understand phenotypic changes induced. Metabolomics one functional level tool employed investigate metabolites with other (metabolism) but also regulatory role through interaction genes, transcripts proteins (e.g. allosteric regulation). Technological developments driving force behind advances scientific knowledge. Recent two analytical platforms mass spectrometry (MS) nuclear magnetic resonance (NMR) spectroscopy have driven forward discipline metabolomics. In this critical review, an introduction metabolites, metabolomes, metabolomics MS NMR will be provided. applications for health-disease continuum, drug efficacy toxicity dietary effects health reviewed. current limitations future goals discussed (374 references).

Introduction The use of innovative technologies is deemed to improve dietary assessment in various research settings. However, their relative merits nutritional epidemiological studies, which require accurate quantitative estimates the usual intake at individual level, still need be evaluated. Objective To report on inventory available for and critically evaluate strengths weaknesses as compared with conventional methodologies (i.e. Food Frequency Questionnaires, food records, 24-hour recalls) used studies. Methods A list currently was identified from English-language journals, using PubMed Web Science. search criteria were principally based date publication (between 1995 2011) pre-defined keywords. Results Six main groups ('Personal Digital Assistant-', 'Mobile-phone-', 'Interactive computer-', 'Web-', 'Camera- tape-recorder-' 'Scan- sensor–based' technologies). Compared Personal Assistant mobile phone devices seem recording through possibility 'real-time' eating events, but validity estimate intakes low moderate. In recalls, there limited knowledge regarding accuracy fully automated approaches; methodological problems, such inaccuracy self-reported portion sizes might more critical than interview-based applications. contrast, measurement errors web-based paper-based Questionnaires are most likely similar, suggesting that underlying methodology unchanged by technology. Conclusions Most new seen have overlapping features methods predominantly epidemiology. Their potential enhance cost- time-effective, less laborious ways data collection higher subject acceptance, though integration studies would additional considerations, study objectives, target population financial resources available. even technologies, inherent bias related will not resolved. More therefore crucial investigate technologies.

The subjective nature of self-reported dietary intake assessment methods presents numerous challenges to obtaining accurate and nutritional status. This limitation can be overcome by the use biomarkers, which are able objectively assess consumption (or exposure) without bias errors. need for biomarkers was addressed Institute Medicine, who recognized lack as a knowledge gap requiring future research. purpose this article is review existing literature on currently available including novel specific foods components, validity, reliability sensitivity markers. revealed several in additional validation research; research also needed produce sensitive, specific, cost-effective noninvasive biomarkers. emerging field metabolomics may help advance development food/nutrient yet advances food metabolome databases needed. availability that estimate components could greatly enhance targeting compliance national recommendations well direct associations with disease outcomes. More necessary refine accounting confounding factors, establish new indicators intake, develop techniques cost-effective, noninvasive, rapid measures
