Depression is apparently the most common psychiatric disease among mood disorders affecting about 10% of adult population. The etiology and pathogenesis depression are still poorly understood. Hence, as for human diseases, animal models can help us understand and, more importantly, may facilitate search therapy. In this review we first describe tests used evaluation depressive-like symptoms in rodents. Then different discuss their strengths weaknesses. These be divided into several categories: genetic models, induced by mental acute chronic stressful situations caused environmental manipulations (i.e., learned helplessness rats/mice), changes brain neuro-transmitters or specific injuries pharmacological tools. spite fact that none completely resembles depression, relevant since they mimic many features observed situation serve a powerful tool study etiology, treatment especially only few patients respond to treatment. Relevance increases also has facets possible etiologies therapies.

Stress is viewed as a state of real or perceived threat to homeostasis, the management which involves endocrine, nervous, and immune systems. These systems work independently interactively part stress response. The scientific literature, spans both animal human studies, contains heterogeneous findings about effects on brain body. This review seeks summarise integrate literature relationships between these systems, examining particularly roles physiological psychosocial stress, hormone cortisol, controlled by hypothalamic-pituitary-adrenal (HPA) axis, cognitive functioning. Health conditions related impaired HPA axis functioning their associated neuropsychiatric symptoms will also be considered. Lastly, this provide suggestions clinical applicability for endocrinologists who are uniquely placed measure outcomes nervous system identify areas intervention.

Despite numerous studies on the neurobiology of depression, etiological and pathophysiological mechanisms this disorder remain poorly understood. A large number animal models tests to evaluate depressive-like behavior have been developed. Chronic unpredictable mild stress (CUMS) is most common frequently used model sucrose preference test (SPT) one for assessing anhedonia. However, not all laboratories can reproduce main effects CUMS, especially when refers a decrease in preference. It also unknown how state anhedonia, assessed by SPT, relates anhedonia patients with depression. We analyzed literature available PubMed database using keywords relevant topic narrative review. hypothesize that poor reproducibility CUMS may be due differences consumption, which influenced such factors as concentration threshold, water food deprivation, animals’ susceptibility stress. believe comparisons between human states should made caution because there are many inconsistencies two, including assessment methods. tried offer some recommendations improve provide framework future research.

As one of most common and severe mental disorders, major depressive disorder (MDD) significantly increases the risks premature death other medical conditions for patients. Neuroinflammation is abnormal immune response in brain, its correlation with MDD receiving increasing attention. has been reported to be involved through distinct neurobiological mechanisms, among which dysregulation neurogenesis dentate gyrus (DG) hippocampus (HPC) The DG two niches adult mammalian neurotrophic factors are fundamental regulators this process. cell types mediating neuroinflammation include microglia, astrocytes, oligodendrocytes, meningeal leukocytes, peripheral cells selectively penetrate blood-brain barrier infiltrate into inflammatory regions. Hallucinations limit widespread therapeutic use of psychedelics as rapidly acting antidepressants. Here we profiled the non-hallucinogenic lysergic acid diethylamide (LSD) analog 2-bromo-LSD (2-Br-LSD) at more than 33 aminergic G protein-coupled receptors (GPCRs). 2-Br-LSD shows partial agonism several GPCRs, including 5-HT2A, and does not induce head-twitch response (HTR) in mice, supporting its classification a 5-HT2A agonist. Unlike LSD, lacks 5-HT2B agonism, an effect linked to cardiac valvulopathy. Additionally, produces weak β-arrestin recruitment internalization vitro tolerance vivo after repeated administration. induces dendritogenesis spinogenesis cultured rat cortical neurons increases active coping behavior blocked by 5-HT2A-selective antagonist volinanserin (M100907). also reverses behavioral effects chronic stress. Abstract Neuropsychiatric disorders, such as major depression, anxiety and post-traumatic stress disorder, tend to be long-term conditions in whose development maintenance are central pathogenic factors. Translational mouse models widely used neuropsychiatric research, exploiting social non-social stressors investigate the mechanisms underlying their detrimental effects. However, most studies focus on short-term consequences of chronic stress, whereas only a few interested course. This is counterintuitive given human that preclinical designed mimic. In this review, we have summarized limited work date effects mice models. First, different presented definition short- vs. sequelae proposed. On basis, behavioral, endocrine, vegetative addressed before examining data cellular molecular alterations brain. Finally, future directions for research discussed.
