The circadian clock is a fundamental timekeeping system that regulates rhythmic biological processes in response to environmental light–dark cycles. In mammals, core genes (CLOCK, BMAL1, PER, and CRY) orchestrate these rhythms through transcriptional–translational feedback loops, influencing various physiological functions, including bone remodeling. Bone homeostasis relies on the coordinated activities of osteoblasts, osteoclasts, osteocytes, with increasing evidence highlighting role regulation maintaining skeletal integrity. Disruptions are linked disorders such as osteoporosis. Posttranslational modifications (PTMs), phosphorylation, acetylation, ubiquitination, serve crucial regulators both mechanisms metabolism. However, specific PTMs integrating timing remodeling remains underexplored. This review examines intersection biology, elucidating their impact cell function homeostasis. Understanding interactions may uncover novel therapeutic targets for diseases associated disruptions.

A molecular clock network is crucial for daily physiology and maintaining organismal health. We examined the interactions importance of intratissue networks in muscle tissue maintenance. In arrhythmic mice showing premature aging, we created a basic module involving central peripheral (muscle) clock. Reconstituting brain-muscle sufficient to preserve fundamental homeostatic functions prevent aging. However, achieving whole requires contributions from other clocks. Mechanistically, acts as gatekeeper, selectively suppressing detrimental signals while integrating important functions. Our research reveals interplay between clocks function underscores impact eating patterns on these interactions.

Circadian clock disruptions, or losses rhythmicity, have been implicated in development diseases, premature ageing, are regarded as health risks. Redox reactions involving reactive oxygen nitrogen species (RONS) regulate physiological functions such cell signalling immune response. However, oxidative stress associated with pathological effects RONS, resulting a loss damaging modifications to important molecules DNA. Direct connections established between rhythms on basis that disruptions can affect redox biology, vice versa, bi-directional relationship. For instance, expression activity antioxidant enzymes (SOD, GPx, CAT) appear follow patterns. Consequently, ability unravel these interactions opened an exciting area biology. Exercise exerts numerous benefits and, potent environmental cue, capacity adjust disrupted systems. In fact, response given exercise stimulus may also exhibit variation. At same time, relationship exercise, scrutinised, whereby it clear exercise-induced RONS elicit both helpful potentially harmful dependent type, intensity, duration exercise. To date, appears emerging interface rhythmicity stress/redox metabolism not explored relation This review aims summarise evidence supporting conceptual link clock, stimuli.
