Abstract Chemodynamic therapy (CDT) based on intracellular Fenton reaction to produce highly cytotoxic reactive oxygen species (ROS) has played an essential role in tumor therapy. However, this still needs be improved by weakly acidic pH and over-expression of glutathione (GSH) microenvironment (TEM), which hinders its future application. Herein, we reported a multifunctional bimetallic composite nanoparticle MnO 2 @GA-Fe@CAI metal polyphenol network (MPN) structure, could reduce endogenous GSH remodeling improve activity. nanoparticles were prepared first @GA-Fe with Fe 3+ as central ion gallic acid (GA) surface ligands the chelation reaction. Then, carbonic anhydrase inhibitor (CAI) was coupled GA form @GA-Fe@CAI. The properties studied, results showed that CAI pH. At same time, deplete Mn 2+ via redox reactions, re-established TME low GSH. In addition, reduced . catalyzed H O high-lever ROS kill cells. Compared , weight volume for xenograft MDA-MB-231 tumor-bearing mice final inhibition rate 58.09 ± 5.77%, showing therapeutic effect well biological safety. The combination of immune checkpoint blockade (ICB) and chemotherapy has shown significant potential in the clinical treatment various cancers. Chemodynamic therapy (CDT) offers an effective in situ activation for cancer treatments. However, the efficiency of CDT is limited by antioxidant system within tumor cells, including antioxidative niacinamide adenine dinucleotide phosphate (NADPH) and glutathione (GSH). Zeolitic imidazolate framework-8 (ZIF-8) widely used therapies due to its tunable structure properties, but ZIF-8 itself lacks application. In order meet application demand CDT, we select Cu2+ ions that can catalyze a Fenton-like reaction 2-nitroimidazole (2-NI) consume NADPH as new inorganic–organic connection components. Based on reasonable coordination mode ZIF-8, nanoplatform with adjustable size constructed. The synthesized Cu-NI NPs are triple responsive hypoxia, GSH, hydrogen peroxide (H2O2), which selectively enhance Cu+/Cu2+-mediated induce apoptosis/ferroptosis. This work will promote diversification structures provide good implications realization enhanced CDT.
