Lipid nanoparticle (LNP) formulations are a proven method for the delivery of nucleic acids gene therapy as exemplified by worldwide rollout LNP-based RNAi therapeutics and mRNA vaccines. However, targeting specific tissues or cells is still major challenge. After LNP administration, LNPs interact with biological fluids (i.e., blood), components which adsorb onto surface forming layer biomolecules termed “biomolecular corona (BMC)” affects stability, biodistribution, tissue tropism. The mechanisms BMC influences tissue- cell-specific remains largely unknown, due to technical challenges in isolating their from complex media. In this study, we present new technique that utilizes magnetic isolate LNP–corona complexes unbound proteins human serum. First, developed formulation, containing >40 superparamagnetic iron oxide nanoparticles (IONPs)/LNP, resulting (IOLNPs) displayed similar particle size morphology loaded acids. We further demonstrated isolation IOLNPs corresponding using separation (MS) system. profile MS system was compared exclusion column chromatography analyzed via mass spectrometry, revealing differences protein abundances. This approach enabled mild versatile its corona, while maintaining structural integrity. identification associated an intact provides insight into interactions fluids.

Inhibition of disease-causing mutations using RNA interference (RNAi) has resulted in clinically approved medicines with additional candidates late stage trials. However, targetable tissues currently preclinical development are limited to liver following systemic intravenous (IV) administration because predictable delivery siRNA non-liver remains an unsolved challenge. Here, evidence durable extrahepatic gene silencing enabled by Selective ORgan Targeting lipid nanoparticles (siRNA SORT LNPs) the kidneys, lungs, and spleen is provided. LNPs excel at dose-dependent tissue-enriched endogenous targets resulting 60%-80% maximal knockdown after a single IV injection up 88% downregulation protein expression mouse lungs two doses. Alzheimer's disease (AD) is a neurological condition currently with 47 million people suffering from it globally. AD might have many reasons such as genetic issues, environmental factors, and Aβ accumulation, which the biomarker of disease. Since primary reason unknown, there no targeted treatment at moment, but ongoing research aims to slow its progression by managing amyloid-beta peptide production rather than symptomatic improvement. phytochemicals been demonstrated possess antioxidant, anti-inflammatory, neuroprotective properties, they may target multiple pathological factors can reduce risk Curcumin, phytochemical found in turmeric known for free radical scavenging an antiamyloid treating AD, has come under investigation. Although low bioavailability limits efficacy, prominent drug delivery system (DDS) desired overcome it. Hence, potency lipid-based nanoparticles encapsulating curcumin (LNPs-CUR) considered this study promising DDS. In vivo studies animal models indicate LNPs-CUR effectively amyloid plaque formation, leading cognitive enhancement reduced toxicity compared CUR. Adjuvanticity and delivery are crucial facets of mRNA vaccine design. In modern vaccines, adjuvant functions integrated into nanoparticles, allowing the co-delivery antigen adjuvants in a unified, all-in-one formulation. this formulation, many vaccines utilize immunostimulating properties carrier components, including lipids polymers, as adjuvants. However, careful design is necessary, excessive adjuvanticity activation improper innate immune signalling can conversely hinder vaccination efficacy trigger adverse effects. also require systems to achieve expression antigen-presenting cells (APCs) within lymphoid organs. Some directly target APCs organs, while others rely on migration draining lymph nodes after taking up vaccines. This review explores current mechanistic understanding these processes ongoing efforts improve safety based understanding.
