The exploration of drugs derived from natural sources holds significant promise in addressing current limitations ovarian cancer (OC) treatments. While previous studies have highlighted the remarkable anti-cancer properties compound β-sitosterol (SIT) across various tumors, its specific role OC treatment remains unexplored. This study aims to investigate anti-tumor activity SIT using vitro and vivo models, delineate potential mechanisms, establish a preclinical theoretical foundation for future clinical trials, thus fostering further research. Utilizing network pharmacology, we pinpoint as promising candidate predict targets pathways. Through series experiments, unveil novel mechanism through which mitigates malignant biological behaviors cells by modulating redox status. Specifically, selectively argininosuccinate synthetase 1 (ASS1), protein markedly overexpressed tissues cells. Inhibiting ASS1, enhances interaction between Nrf2 Keap1, instigating ubiquitin-dependent degradation Nrf2, subsequently diminishing transcriptional activation downstream antioxidant genes HO-1 NQO1. interruption program results substantial accumulation reactive oxygen species (ROS) This, turn, upregulates PTEN, exerting negative regulation on phosphorylation AKT. suppression AKT signaling disrupted pathways associated with cell cycle, survival, apoptosis, migration, invasion, ultimately culminating death Our research uncovers new mechanisms against OC, contributing existing knowledge effects products context OC. Additionally, this unveils ASS1 regulating Nrf2-mediated governing homeostasis providing deeper understanding complex disease.

Artemisia campestris subsp. (tuguft) is a medicinal plant traditionally used in Algerian medicine. This study investigates the chemical composition and bioactivity of its essential oil (ACEO). Gas chromatography-mass spectrometry (GC-MS) analysis identified key compounds, including linalyl acetate (2.92%), geranyl (2.45%), eucalyptol (1.38%). ACEO demonstrated significant antioxidant activity, with IC50 values 11.09 μg/mL (DPPH), 15.81 (FRAP), 22.70 (β-carotene). It also enhanced peroxidase activity by 82.67 U/g. The anti-inflammatory effects were confirmed an 18.87 μg/mL. Notably, silico molecular docking revealed that 3-cyclopentyl-N-(2-(3,4-dimethoxyphenyl)ethyl) exhibits strong binding affinity to phosphoinositide 3-kinase gamma, target for pancreatic cancer therapy, suggesting potential anticancer activity. These findings underscore therapeutic ACEO, highlighting antioxidant, anti-inflammatory, properties.

Abstract Background Glioblastoma (GBM) is the most common primary malignant tumor in central nervous system. Paclitaxel (PTX) a well‐established and highly effective anti‐cancer drug for peripheral solid tumors. However, application of PTX GBM hindered by several limitations, including poor water solubility, restricted entry across blood–brain barrier (BBB), enhanced excretion efflux transporters. P‐glycoprotein (P‐gp) crucial transporter that abundantly present cerebral vascular endothelial cells cells. It plays significant role exocytosis within tissues. Methods Recently, we have developed novel technique creating self‐assembled nanoparticles utilizing range natural bioactive molecules. These can encapsulate insoluble drugs effectively cross BBB. In additional, revealed certain potential to act as P‐gp inhibitors, thereby reducing PTX. this study, conducted screening molecular identify those inhibit function Results Among candidates, identified ursolic acid (UA NPs) inhibitors. Furthermore, prepared co‐assembled UA NPs embedded with paclitaxel, referred UA‐PTX NPs. Our results demonstrate enhance blood concentration PTX, facilitate its into BBB, P‐gp, resulting decrease This discovery addressed above three issues associated use glioma treatment.
