Aim: The fixed-dose combination of moxifloxacin (MOXI) and ketorolac tromethamine (KTR) is widely used for the treatment bacterial keratitis. Thus, a new LC–MS/MS method was developed to determine MOXI KTR in lacrimal fluid. Methods: Bioanalysis performed using Shimadzu 8050 electrospray ionization-positive mode validated per US FDA guidelines. Isocratic separation with Waters Symmetry C18 column methanol 0.1% formic acid containing deionized water (85:15, v/v). For this research, we performed a 2D-QSAR analysis on set of 34 molecules derived from dimedone with inhibitory activity against human colon cancer (HT-29). The investigation incorporated principal component (PCA), multiple linear regression (MLR), non-linear (MNLR), and artificial neural network (ANN). evaluations the QSAR models demonstrated high predictive power (R2MLR=0.884; R2CV (MLR)=0.86), (R2MNLR=0.810; (MNLR)=0.879), (R2ANN=0.9; R2CV(ANN)=0.89). reliability was validated through internal, external, Y-randomization, validation domain applicability assessments. Utilizing model predictions, designed four new molecular structures that exhibited superior HT-29 cell line compared to previously tested compounds.
