Camptothecin (CPT) is a highly cytotoxic molecule with excellent antitumor activity against various cancers. However, its clinical application severely limited by poor water solubility, easy inactivation, and severe toxicity. Structural modifications nanoformulations represent two crucial avenues for camptothecin's development. the potential further structural limited, camptothecin nanoparticles fabricated via physical loading have drawbacks of low drug leakage. Prodrug-based CPT shown unique advantages, including increased loading, reduced burst release, improved bioavailability, minimal toxic side effects. Stimulus-responsive nano-prodrugs that respond to endogenous or exogenous stimuli introducing activatable linkers achieve spatiotemporally responsive release at tumor site. This review comprehensively summarizes latest research advances in stimulus-responsive nano-prodrugs, preparation strategies, mechanisms, their applications cancer therapy. Special focus placed on mechanisms characteristics treatment. Furthermore, prodrugs are discussed. Finally, challenges future directions be valuable readers engaged prodrug expected.

Bioactive compounds represent a broad class of dietary metabolites derived from fruits and vegetables, such as polyphenols, carotenoids glucosinolates with potential for cancer prevention. Curcumin, resveratrol, quercetin, β-carotene have been the most widely applied bioactive in chemoprevention. Lately, many approaches to encapsulating components nano-delivery systems improved biomolecules' stability targeted delivery. In this review, we critically analyze compounds, including polymeric nanoparticles (NPs), solid lipid (SLN), nanostructured carriers (NLC), liposomes, niosomes, nanoemulsions (NEs) use therapy. Immunotherapy as an alternative treatment strategy for B-cell lymphoma is undesirable because of tumor heterogeneity and immune surveillance. Spermidine (SPM), a regulator the microenvironment (TME), can facilitate release damage-associated molecular patterns (DAMPs) from cancer cells, promote recognition, thus alleviate surveillance in TME. Hence, this work, self-assembled spermidine-based metal-immunopeptide nanocomplexes (APP-Fe NCs; APP anti-programmed death ligand-1 peptide) with pH-responsive kinetics were prepared via flash nanocomplexation (FNC) technique based on noncovalent interaction between APP-SPM-dextran (DEX) sodium tripolyphosphate (TPP) coordination Fe3+ TPP. An vitro study suggested that APP-Fe NCs effectively induce strong oxidative stress mitochondrial dysfunction subsequently lead to ferroptosis cells by interfering homeostasis cells. Further investigation mice models demonstrated inhibited growth liver metastasis lymphomas. Mechanistically, triggering tissues, these spermidine-containing efficiently facilitated DAMPs ultimately reshaped TME enhance immunotherapy efficacy lymphoma. Immunotherapy gains increasing focus in treating triple-negative breast cancer (TNBC), while its efficacy is greatly restricted owing to low tumor immunogenicity and immunosuppressive microenvironment (ITM). Herein, a LyP-1 chondroitin sulfate (CS) dual-modified liposome co-loaded with paclitaxel (PTX) cryptotanshinone (CTS), namely CS/LyP-1-PC Lip, engineered for TNBC chemoimmunotherapy via induction of immunogenic cell death (ICD) inhibition signal transducer activator transcript-3 (STAT3) activation. Lip enhances cellular uptake through p32 CD44 dual receptor-mediated endocytosis. Within the tumor, CS layer continuously detached by hyaluronidase release drugs. Subsequently, CTS sensitizes cytotoxicity PTX 4T1 cells. induces ICD cells facilitates infiltration cytotoxic T lymphocyte provoke immune response. Meanwhile, concomitant delivery inhibits STAT3 activation decrease regulatory cell, M2-type tumor-associated macrophage, myeloid-derived suppressor thus reversing ITM. Markedly, dual-targeting shows superior anti-tumor subcutaneous mice significant lung metastasis suppression model. Overall, this work offers feasible combination regimen promising nanoplatform development chemoimmunotherapy.
