As the most widely used herbal medicine in human history and a major defence hormone plants against broad spectrum of pathogens abiotic stresses, salicylic acid (SA) has attracted research interest. With applications modern technologies over past 30 years, studies effects SA on plant growth, development, have revealed many new frontiers continue to deliver surprises. In this review, we provide an update recent advances our understanding metabolism, perception, signal transduction mechanisms immunity. An overarching theme emerges that executes its functions through intricate regulation at multiple steps: biosynthesis is regulated both locally systemically, while perception occurs cellular targets, including metabolic enzymes, redox regulators, transcription cofactors, and, recently, RNA-binding protein. Moreover, orchestrates complex series post-translational modifications downstream signaling components promotes formation biomolecular condensates function as signalling hubs. also impacts wider crosstalk with other hormones. Looking into future, propose areas for exploration functions, which will undoubtedly uncover more surprises years come.

Pathogen infection is a dynamic process. Here, we employ single-cell transcriptomics to investigate plant response heterogeneity. By generating an Arabidopsis thaliana leaf atlas encompassing 95,040 cells during by fungal pathogen, Colletotrichum higginsianum, unveil cell-type-specific gene expression, notably enrichment of intracellular immune receptors in vasculature cells. Trajectory inference identifies that had different interactions with the invading fungus. This analysis divulges transcriptional reprogramming abscisic acid signaling specifically occurring guard cells, which consistent stomatal closure dependent on direct contact Furthermore, plasticity genes involved glucosinolate biosynthesis at sites, emphasizing contribution epidermis-expressed MYB122 disease resistance. work underscores spatially dynamic, responses pathogen and provides valuable resource supports in-depth investigations plant-pathogen interactions.

Abstract Plant diseases are a constant and serious threat to agriculture ecological biodiversity. Plants possess sophisticated innate immunity system capable of detecting responding pathogen infection prevent disease. Our understanding this has grown enormously over the past century. Early genetic descriptions plant disease resistance virulence were embodied in gene-for-gene hypothesis, while physiological studies identified pathogen-derived elicitors that could trigger defense responses cells tissues. Molecular these phenomena have now coalesced into an integrated model involving cell surface intracellular detection specific molecules proteins culminating induction various cellular responses. Extracellular receptors engage distinct signaling processes but converge on many similar outputs with substantial evidence for integration pathways interdependent networks controlling outcomes. Abstract Plants lack specialized and mobile immune cells. Consequently, any cell type that encounters pathogens must mount responses communicate with surrounding cells for successful defence. However, the diversity, spatial organization function of cellular states in pathogen-infected plants are poorly understood 1 . Here we infect Arabidopsis thaliana leaves bacterial trigger or supress integrate time-resolved single-cell transcriptomic, epigenomic transcriptomic data to identify states. We describe cell-state-specific gene-regulatory logic involves transcription factors, putative cis -regulatory elements target genes associated disease immunity. show a rare population emerges at nexus immune-active hotspots, which designate as primary responder (PRIMER) PRIMER have non-canonical signatures, exemplified by expression genome accessibility previously uncharacterized factor, GT-3A, contributes plant immunity against pathogens. surrounded another state (bystander) activates long-distance cell-to-cell signalling. Priming reflects the capacity of plants to memorise environmental stress experience and improve their response recurring stress. Epigenetic modifications in DNA associated histone proteins may carry short-term long-term memory same plant or mediate transgenerational effects, but evidence is still largely circumstantial. New experimental tools now enable scientists perform targeted manipulations that either prevent generate a particular epigenetic modification location genome. Such 'reverse epigenetics' approaches allow for interrogation causality between individual priming-induced role altering gene expression performance under Furthermore, combining site-directed manipulation with conditional cell-type specific promoters creates novel opportunities test engineer spatiotemporal patterns priming.
