Lipophosphoglycan (LPG) is a macromolecule on the surface of Leishmania spp. parasites. The biochemical structure LPG varies throughout parasites' life cycle between proliferative (procyclic) and infective (metacyclic) stages, as well species strains. Here, we outline protocol for growing parasites in vitro to harvest LPG. We describe steps parasite differentiation extraction purification. has applications medical research, such trained immunity immunotherapy cancer. Leishmaniasis is a collective term for several tropical, neglected diseases caused by protozoans of the species Leishmania, 20 which causing disease in humans ranging from localised self-healing lesions to chronic manifestations affect skin or inner organs. Although millions infections are accounted annually, treatment options scarce and limited medication associated with heavy side-effects increasing antibiotic resistance. Case studies point towards immunotherapy as effective alternative relying on immunomodulatory properties e.g., Bacillus Calmette-Guérin vaccine. Leishmania parasites also known modulate immune system, yet underlying macromolecules surface molecules remain widely under characterised. With this short review, we aim provide complete summary existing literature describing one most expressed molecule spp, lipophosphoglycan (LPG), shows great variability between different lifecycle stages spp. Complete characterisation LPG may aid improve development vaccination strategies, open new avenues exploit unrelated conditions.

Methicillin-resistant Staphylococcus aureus (MRSA) represents a major global health threat due to its resistance conventional antibiotics. The commensal microbiota maintains symbiotic relationship with the host, playing essential roles in metabolism, energy regulation, immune modulation, and pathogen control. Mammals harbor wide range of bacteria capable producing unique metabolites potential therapeutic properties. This study demonstrated that M28 family peptidase (M28), derived from Peribacillus frigoritolerans (P. f), provided protective effects against MRSA-induced pneumonia. enhanced phagocytosis bactericidal activity macrophages by inducing trained immunity. RNA sequencing metabolomic analyses identified CFB-C3a-C3aR-HIF-1α axis-mediated phosphatidylcholine accumulation as key mechanism for M28-induced Phosphatidylcholine, like M28, also induced To enhance M28-mediated potential, it was encapsulated liposomes (M28-LNPs), which exhibited superior immune-stimulating properties compared alone. In vivo experiments revealed M28-LNPs significantly reduced bacterial loads lung damage following MRSA infection, protection Klebsiella pneumoniae Candida albicans. We first confirmed link between complement activation immunity, offering valuable insights into treatment prevention complement-related autoimmune diseases.

The current understanding of innate immune memory encompasses both trained immunity and tolerance, where cells can exhibit enhanced responsiveness or paralysis upon subsequent stimuli, respectively. Various agents induce either these responses, including β-glucan, Leishmania, BCG LPS. is a clinically approved immunotherapy for bladder cancer BCG-induced important in driving anti-tumor adaptive immunity. Salmonella also shows promise treatment, eliciting potent but with transitory effects. This led us to investigate whether LVR01, like BCG, triggers its impact on responses. Herein, we report that induces an response bone marrow cells, characterized by robust cytokine second stimulus, fashion resembles Coherently that, administration tumor implanted later time, resulting slow growth extended survival. However, vitro stimulation human monocytes murine bone-marrow derived myeloid-enriched results decreased production cytokines resembling paralysis. Overall, our suggest LVR01 responses memory, as well monocytes. These two antagonistic effects could be the basis effect treatment further investigation phenomena shed light how improve Salmonella-based immunotherapies cancer.

Trained immunity (TI) is functional memory displayed by innate immune cells (IICs). TI facilitates rapid, non-specific responses to pathogens upon secondary challenge. It driven immunological signaling and metabolic rewriting via epigenetic alteration, triggered recognition of certain stimuli. Recently, checkpoint inhibitors have come into common use in clinical oncology settings, genetically engineered cytotoxic T comprise a potent cancer treatment strategy. However, the contributions tumor microenvironment (TME) are only beginning be uncovered. Accumulating evidence that various microorganisms vaccines convey tumoricidal ability suggest may become useful anti-cancer tool. The expected roles therapy 1) promotion proinflammatory cytokine section, 2) enhancement phagocytosis, 3) quick expansion recruitment cancer-specific TME through neoantigen presentation, 4) reversal immunosuppression TME, 5) removal associated with carcinogenesis or development. Medium- long-term durability reduce risk Recent findings on usher new aspirations for treatment.
