Abstract Escitalopram (ETP) has poor oral bioavailability due to its low water solubility, hence the goal of this work was design and optimize a self-nano-emulsifying drug delivery system (SNEDDS). Using results investigations on solubility emulsification, pseudo-ternary phase diagram produced. The three main ingredients chosen for formulation were polyethylene glycol 400 (co-surfactant), tween 80 (surfactant), geranium oil (lipid). ETP-SNEDDS evaluated size particles surface charge. Fourier transforms infrared spectroscopy (FTIR), differential scanning calorimetry (DSC) thermogravimetric analysis (TGA) used evaluate chemical compatibility thermal stability. Ex-vivo permeability, in vitro digestion, dissolution carried out compared with reference tablets. ETP-loaded SNEDDS comparison control Wistar rats (n = 6). Vesicants are chemical warfare agents (CWAs) capable of causing severe skin damage and systemic toxicity. Melatonin, known for its anti-inflammatory antioxidant properties, can mitigate the effects these agents. Self-nano-emulsifying drug delivery systems (SNEDDS) containing a high melatonin concentration (5 %, 50 mg/g) were optimized using quality-by-design approach from biocompatible, non-irritant excipients with particle size about 100 nm. The melatonin-loaded SNEDDS showed 43-fold greater permeability than conventional cream. Chemical stability at ambient temperature (25 °C) was maintained one year. preparation optimised simple mixing method compared to microfluidic micromixers. Mixing successfully achieved 3D-printed (fused deposition modeling or stereolithography) T-shaped toroidal chip (with channel geometry by computational fluid dynamics), resulting in scalable, continuous process first time substantial reduction other approaches. Atopic dermatitis (AD) is the most common form of eczema and burdensome skin disease globally, affecting nearly 223 million. A major AD predisposition genetic susceptibility, barrier integrity cell-mediated immunity. Manifesting as red, dry, itchy skin, basic treatment involves hydration with emollients. Despite their effectiveness, poor patient compliance remains a drawback. In severe cases, medicated emollients are used, but carry risks, including thinning, immunosuppression. Hence, hydrogels have emerged promising alternative for management based on ability to improve hydration, attributed hydrophilicity high water retention capacity. Moreover, researchers loaded various compounds management; they also hold potential reduce systemic side effects commercial drugs by enhancing dermal retention. Hydrogels address challenges non-greasy texture reduced application frequency. Their appeal stems from versatility, can be fabricated varying polymers.
