Abstract Background Cathepsin S (CTSS) is a cysteine protease that played diverse roles in immunity, tumor metastasis, aging and other pathological alterations. At the cellular level, increased CTSS levels have been associated with secretion of pro-inflammatory cytokines disrupted homeostasis Ca 2+ flux. Once was suppressed, elevated anti-inflammatory changes influx were observed. These findings inspired us to explore potential role on cognitive functions. Methods We conducted classic Y-maze Barnes Maze tests assess spatial working memory Ctss −/− mice, + / mice injected inhibitor (RJW-58). Ex vivo analyses including long-term potentiation (LTP), Golgi staining, immunofluorescence staining sectioned whole brain tissues obtained from experimental animals conducted. Furthermore, molecular studies carried out using cultured HT-22 cell line primary cortical neurons treated RJW-58 comprehensively gene protein expressions. Results Our reported targeting cathepsin yields improvements function, enhancing both behavior models. showed synaptic complexity. Microarray analysis demonstrated brain-derived neurotrophic factor (BDNF) upregulated when knocked down by siRNA. Moreover, pharmacological blockade enzymatic activity promoted BDNF expression dose- time-dependent manner. Notably, inhibition neurogenesis murine dentate gyrus. results suggested promising modulation enhancement neurogenesis. Conclusion suggest critical regulation function modulating influx, leading enhanced activation BDNF/TrkB axis. study may provide novel strategy for improving CTSS.
