Abstract Gastric ulcer is a common peptic that affects human health and life quality seriously. As anti‐gastric drugs usually cause side‐effects, polysaccharides may be the potential alternatives because of better effectiveness less toxicity. Although activities have been widely reported, mechanisms not yet well‐disclosed. In this study, network pharmacology analysis was performed to explore against gastric ulcer, results were validated by molecular docking. Results indicated β‐glucan, arabinogalactan, xylan, arabinan key structures, ABL1, AKT1, androgen receptor, epidermal growth factor v‐Ha‐ras Harvey rat sarcoma viral oncogene homolog, HSP90AA1, mitogen‐activated protein kinase 8 (MAPK8), MAPK14, NOS2, PIK3R1, RAC1, ras homolog gene family member A, proto‐oncogene tyrosine‐protein Src core targets for in treating ulcer. Polysaccharides influences on 1958 GO items 199 KEGG pathways, their are related MAPK, Ras, PI3K‐Akt, vascular endothelial factor, prolactin, FoxO Rap1 signaling etc. Molecular docking validation showed credible, interactions between polysaccharide structures observed. This study contributes understanding provides references future activity screening mechanism research

ABSTRACT Scope: Gallic acid (GA) is recognized for its purported antiinflammatory properties. GA has been demonstrated to prevent and alleviate the symptoms of chronic colitis through modulation gut microbiota, improvement intestinal barrier, reduction inflammation.Methods results: In order determine mechanism by which exerts protective effect against colitis, mice were induced dextran sulfate sodium (DSS). The in disease activity index 25% decrease colon tissue damage indicated that 36 days intervention alleviated DSS‐induced symptoms. was observed mitigate weight loss 2.5% shortening 17.3%, diminish expression pivotal proteins within TLR4/nuclear factor κB (NF‐κB) signaling cascades, consequently lowering generation inflammatory cytokines. Furthermore, effectively corrected microbiota imbalance, increased content short‐chain fatty acids (SCFAs), turn suppressed inflammation, enhanced tight junction protein expression, thereby strengthening barrier. Conclusion: capacity enhance efficacy a multifaceted mechanism, influencing barrier function, processes. findings highlight potential as preventative strategy colitis.

This study aimed to investigate the effects of Lyophyllum decastes β-glucan (LDBG) in preventing high-fat diet (HFD)-induced obesity male C57BL/6 mice, focusing on fecal gut microbiota (GM) and plasma metabolomics. LDBG treatment led a reduction body weight an improvement dyslipidemia HFD-fed mice. Additionally, decreased pro-inflammatory factors (TNF-α, IL-1β, IL-6) liver abdominal adipose tissue lowered lipopolysaccharide leptin levels plasma. also up-regulated mRNA intestinal epithelial tight junction proteins, such as Occludin, Claudin-1, zonula occludens-1 colon tissue. Furthermore, 16S rRNA gene sequencing data suggested that reduced relative abundance Faecalibaculum Ileibacterium while increasing Bifidobacterium Oscillospiraceae. Untargeted metabolomics showed significantly promoted Tryptophan metabolism, Glycerophospholipid Citrate cycle, closely correlating with GM changes, particularly Oscillospiraceae genera. demonstrates potential prebiotic prevention by regulating metabolite pathways.
