Dendrobium officinale (DEN) is recognized as a kind of functional food that can effectively ameliorate endocrine and metabolic disruptions. This study delved into the pharmacological mechanism DEN on hepatic lipotoxicity associated with Type II diabetes mellitus (T2DM). In vivo experiments db/db mice indicated treatment notably enhanced liver function, decreased blood lipid levels, improved insulin sensitivity. Non-targeted metabolomics analysis revealed significantly ameliorated metabolism pathways, including lipoic acid, linoleic bile secretion, alanine/aspartate/glutamate metabolism, well taurine hypotaurine metabolism. Transcriptomics demonstrated could modulate expression genes such Cpt1b, Scd1, G6pc2, Fos, Adrb2, Atp2a1, Ppp1r1b, Cyp7a1. Furthermore, Proteomics beneficial effect was linked to pathways like AMPK PPAR signaling. The integrative multi-omics PPAR-RXR signaling critical therapeutic T2DM-induced fatty liver. Additionally, in vitro AML-12 cells confirmed counteract PA-induced accumulation by activating pathway. Overall, these findings suggested exhibited potential mitigate lipo-toxicity manage imbalances T2DM.

Polygonatum sibiricum polysaccharide (PSP) was extracted and purified from raw material obtained P. sibiricum. The structural features of PSP were investigated by Congo red, circular dichroism spectrum, high-performance gel permeation chromatography, scanning electron microscope, atomic force ultraviolet spectroscopy, Fourier transform infrared spectroscopy analysis. In vitro simulations conducted to investigate the kinetics enzyme inhibition. Moreover, a type II diabetes mouse model (T2DM) with streptozotocin-induced insulin resistance established, indexes lipid quadruple, index, oral glucose tolerance (OGTT), organ pancreatic morphology mice measured. results showed that mainly consists monosaccharides, such as mannose, glucose, galactose, xylose, arabinose. It also has β-glycosidic bond pyranose ring an irregular reticulated aggregated structure triple helix. inhibition assays revealed acts reversible competitive inhibitor α-glucosidase α-amylase. Furthermore, found reduce increase OGTT serum levels, decrease free fatty acid content improve metabolism, lower glycated protein enhance metabolism in T2DM mice, thereby leading reduction blood concentration. Additionally, exhibited reparative effects on damaged liver tissue cells mice. experiment provide preliminary basis for therapeutic mechanism about theoretical reference application food pharmaceutical development.
