Abstract Psoriasis is a multifactorial immune-mediated inflammatory disease. Its pathogenesis involves abnormal accumulation of neutrophils and T-cell related abnormalities. Pyroptosis type regulated cell death associated with innate immunity, but its role in psoriasis unclear. In this study, we found that gasdermin D (Gsdmd) higher human psoriatic skin than normal skin, imiquimod-induced psoriasis-like mouse the expression Gsdmd was most significantly altered Il1b also mainly expressed neutrophils. Immunohistochemical staining serial sections lesions from patients healthy control showed GSDMD lesion, especially deficiency mitigates inflammation mice. contributes to inflammation, while depletion attenuates development reduces release cytokines. We neutrophil pyroptosis involved which provides new insights into treatment by targeting pyroptosis.

Results antagonized proliferation, inflammatory response, NLRP3/caspase‐1/Gasdermin‐D (GSDMD)‐mediated pyroptosis M5‐challenged cells. Besides, NLRP3 elevation partially abrogated effects M5‐induced NLRP3/caspase‐1/GSDMD‐mediated Conclusion In word, might exert anti‐proliferation, anti‐inflammatory anti‐pyroptosis model blockade NLRP3/Caspase‐1/GSDMD pathway be recognized as potential mechanism underlying psoriasis, suggesting prospective anti‐psoriasis drug.
