Abstract Tumor-associated inflammation plays a vital role in cancer progression. Among the various stromal cells, cancer-associated fibroblasts are promising targets for therapy. Several reports have indicated potent anti-inflammatory effects attributed to Curcumin. This study aimed investigate whether inhibiting inflammatory function of (CAFs) with Curcumin can restore anticancer immune responses. CAFs were isolated from breast tissues, treated Curcumin, and co-cultured patients' PBMCs evaluate gene expression cytokine production alterations. Blood tumor tissue samples obtained 12 patients stage II/III invasive ductal carcinoma. Fibroblast Activation Protein (FAP) + extracted tissue, 10 μM corresponding PBMCs. The smooth muscle actin-alpha (α-SMA), Cyclooxygenase-2(COX-2), PGE2, cell cytokines evaluated using Real-Time PCR ELISA, respectively. Analyzes showed that treatment decreased genes α-SMA COX-2 PGE2 CAFs. In Curcumin-treated CAFs, FoxP3 along TGF-β, IL-10, IL-4. An increase IFN-γ was observed followed by increased T-bet expression. According our results, could reprogram pro-tumor phenotype anti-tumor Thus, as component microenvironment, suitable target combination immunotherapies cancer.

Abstract Anti‐inflammatory compounds, delivered as a payload to the gastrointestinal tract (GIT) by carriers, still cannot treat inflammatory bowel diseases without avoiding side effects. Here, we developed payload‐free protein nanoparticles (PNPs) that crossed GIT retain in colon and colitis restoring intestinal barrier integrity modulating gut microbiome metabolome. Specifically, PNPs, orally administered mice with acute colitis, reached within three hours. Consequently, PNPs improve microbiota dysbiosis reverse metabolism balance, suppressing expression of tumor‐necrosis factor α toll‐like receptor 4 restores integrity. then ameliorated inflammation attenuated exerting probiotic effects on microbiota, treating week more effectively than clinically often used 5‐aminosalicylic acid causing undesired Such represent safe, sustainable, cost‐effective therapeutics for metabolic eliminating microbial metabolomic imbalance.
