Transdermal delivery provides numerous benefits over conventional routes of administration. However, this strategy is generally limited to a few molecules with specific physicochemical properties (low molecular weight, high potency, and moderate lipophilicity) due the barrier function stratum corneum layer. Researchers have developed several physical enhancement techniques expand applications transdermal field; among these, microneedle technology has recently emerged as promising platform deliver therapeutic agents any size into across skin. Typically, hydrophilic biomolecules cannot penetrate skin by passive diffusion. Microneedle insertion disrupts integrity compromises its protective function, thus creating pathways (microchannels) for enhanced permeation macromolecules. Microneedles not only improve stability but also enhance various biomolecules. Academic institutions industrial companies invested substantial resources in development systems biopharmaceutical delivery. This review article summarizes most recent research provide comprehensive discussion about microneedle-mediated macromolecules, covering topics from introduction skin, delivery, microneedles, biopharmaceuticals (current status, administration, issues), different types, clinical trials, safety acceptability manufacturing regulatory issues, future technology.

Microneedles (MNs) can be used for the topical treatment of skin disorders as they directly deliver therapeutics to site lesions, resulting in increased therapeutic efficacy while having minimum side effects. MNs are different kinds (e.g., small molecules, macromolecules, nanomedicines, living cells, bacteria, and exosomes) treating various disorders, including superficial tumors, wounds, infections, inflammatory diseases, abnormal appearance. The improved by integrating advantages multiple perform combination therapy. Through careful designing, further modified with biomimetic structures responsive drug release from internal external stimuli enhance transdermal delivery efficiency robust outcomes. Some studies have proposed use drug-free a promising mechanotherapeutic strategy promote wound healing, scar removal, hair regeneration via mechanical communication pathway. Although several advantages, practical application suffers problems related industrial manufacture clinical evaluation, making it difficult translation. In this study, we summarized applications, emerging challenges, developmental prospects provide information on ways advance

Abstract Rheumatoid arthritis (RA) is a debilitating autoimmune disease that causes chronic pain and serious complications, presenting significant challenge to treat. Promising approaches for treating RA involve signaling pathways modulation targeted therapy. To this end, multifunctional nanosystem, TPC‐U@HAT, has been designed therapy, featuring multitargeting, dual‐stimuli response, on‐demand drug release capabilities. TPC‐U@HAT composed of probe/prodrug TPC, JAK1 kinase inhibitor upadacitinib, the carrier HAT. TPC an aggregation‐induced emission (AIE)‐active NIR‐II chromophore TPY NF‐κB/NLRP3 caffeic acid phenethyl ester (CAPE), connected via boronic bond which serves as reactive‐oxygen‐species‐responsive linker. The carrier, HAT, created by grafting bone‐targeting alendronate hydrophobic tocopheryl succinate onto hyaluronic chains, can encapsulate upadacitinib form TPC‐U@HAT. Upon intravenous injection into mice, accumulates at inflamed lesions through both active passive targeting, overexpressed hyaluronidase H 2 O therein cleave polymer chains boronate bonds, respectively. This generates AIE‐active detection therapeutic evaluation optoacoustic imaging fluorescent concomitantly releases CAPE exert efficacious therapy inhibiting JAK‐STAT pathways.

The skin is considered the largest and most accessible organ in human body, allows use of noninvasive efficient strategies for drug administration, such as transdermal delivery system (TDDS). TDDSs are systems or patches, with ability purpose to deliver effective therapeutic doses drugs through skin. Regarding specific interaction between hydrogels (HG) microneedles (MNs), we seek find out how this combination would be applied context delivery, detail some possible advantages methods used. Depending on components belonging HG matrix, can obtain essential characteristics that make hydrogels–microneedles (HG–MNs) very advantageous, response external stimuli, among others. Based multiple provided by HGMNs depicted work, it unique properties include controlled, sustained, localized release, well possibility a synergistic association formulation more than one bioactive component. In conclusion, based HG–MNs offer many biomedical field, bringing light new technological safe improving pharmacokinetics pharmacodynamics treatment perspectives.
