Due to oral nano-delivery systems for the treatment of inflammatory bowel disease (IBD) are often failed accumulated colonic site and could not achieve controlled drug release, it's urgent develop a microenvironment responsive delivery improve therapy efficacy. Inflammation at IBD is mainly mediated by macrophages, which key effector cells. Excessive inflammation leads oxidative stress intestinal mucosal damage. The use curcumin (CUR) emodin (EMO) together promising due their respective anti-inflammatory repair effects. In view pH gradient environment gastrointestinal tract, here we prepared pH-responsive sodium alginate (SA) hydrogel-coated nanoemulsions co-deliver CUR EMO (CUR/EMO NE@SA) release specifically target macrophages colon. In this study, CUR/EMO NE@SA was successfully developed, in NE loaded chitosan further crosslinked with alginate. had property colon. preparation significantly alleviate colon decreasing TNF-α IL-6 expression, increasing IL-10 scavenging reactive oxygen species ameliorating restoration tight junction protein expression. Furthermore, revealed molecular mechanism treatment, might synergic inhibition NF-κB pro-inflammatory microenvironment. Our study provides new strategy via synergically inhibiting inflammatory, repairing clearing ROS pH-sensitive hydrogel-encapsulated nanoemulsion system, be developed other chronic treatment. It's suggested that nanoemulsion-based codelivery combinatorial platform IBD.

Rheumatoid arthritis (RA) is a chronic inflammatory joint disease accompanied by energy depletion and accumulation of reactive oxygen species (ROS). Inorganic nanoparticles (NPs) offer great promise for the treatment RA because they mostly have functions beyond being drug carriers. However, conventional nanomaterials become coated with protein corona (PC) or lose their cargo prematurely in vivo, reducing therapeutic efficacy. To avoid these problems, we loaded methotrexate (MTX) into hollow structured manganese dioxide (H-MnO2 NPs), then them 'pseudo-corona' human serum albumin (HSA) at physiological concentrations to obtain HSA-MnO2@MTX NPs. Efficacy MTX, MnO2@MTX, NPs was compared vitro vivo. Compared HSA-coated were taken up better lipopolysaccharide-activated RAW264.7 more effective lowering levels pro-inflammatory cytokines preventing ROS accumulation. also efficient blocking proliferation migration fibroblast-like synoviocytes from rats collagen-induced arthritis. In this rat model, showed biodistribution than other treatments, specifically targeting ankle joint. Furthermore, reduced swelling paw, regulated cytokine production, limited cartilage degradation signs inflammation. These results establish potential against RA.
