The regeneration of adult skeletal muscle after injury is primarily initiated by satellite cells (SCs), but the regulatory mechanisms committed to myogenic differentiation remain poorly explored. Small molecular selenoprotein K (SelK) plays crucial roles in modulation endoplasmic reticulum (ER) stress and against oxidative stress. Here, we first showed that SelK expression activated during both vivo vitro. Meanwhile, loss delayed regeneration, inhibited development myoblasts into myotubes, was accompanied reduced factors (MRFs). Moreover, ER stress, intracellular reactive oxygen species (ROS), autophagy apoptosis under myogenesis induction were more severe SelK-deficient mice than corresponding control groups. Supplementation with specific inhibitors alleviate excessive or partly rescued potential formation myotubes. Notably, demonstrated Self-mediated regulation cellular redox status derived from its subsequent effects on Together, our results suggest protects damage a regulator myogenesis.

Microplastics (MPs) generally refer to the plastic fragments or particles smaller than 5 mm in diameter, which are closely concerned due their widespread presence environment. Recent studies have shown that MPs a serious threat on reproductive health of organisms. Pigs often selected as model animals because high similarity human tissues and organs. However, there no reports effects mechanisms exposure swine germ cells. In present study, we established testis (ST) cell models exposed 250, 500, 1000 μg/ml polystyrene microplastics (PS-MPs, 1-10 μm), respectively. The findings revealed PS-MPs reduced viability dose-dependently. Acridine orange/ethidium bromide staining flow cytometry results indicated occurrence apoptosis necrosis ST cells under exposure, expression changes relevant marker genes (B-cell lymphoma-2, Bcl-2 Associated X, Caspase-3, Caspase-9, Receptor-interacting protein kinase 1, 3, Mixed lineage domain-like, Caspase-8) were clarified via quantitative real-time PCR western blot. Further mechanistic found treatment induced excessive intracellular reactive oxygen species (ROS) production, promoted phosphorylation mitogen-activated (MAPK) pathway-related (P38, c-Jun N-terminal kinase, extracellular regulated kinases) activated downstream gene hypoxia-inducible factor (HIF1α). conclusion, our study suggests causes necroptosis ROS/MAPK/HIF1α signaling pathway.
