Arsenic (As) and Cadmium (Cd) are toxic to rice plants. However, selenium (Se) has the potential regulate As Cd toxicity. The present study aimed evaluate co-exposure As5+ Se6+ species in two cultivars, BRS Pampa EPAGRI 108. plants were divided into six groups cultivated until complete maturation of grains, under greenhouse conditions. Regarding total inorganic (i-As) accumulation highest concentrations found for Pampa. For Se, 108 presented concentration organic Se (i-Se o-Se). exposure assessments showed that biofortification can mitigate and, consequently, risk toxicity grains human consumption. combined effect could represent an alternative biofortify this food a safe way with higher percentage bioavailable Se. Although is able plants, we different cultivars same growing conditions may responses exposure.

Abstract Intervertebral disc degeneration (IVDD) is one of the most prevalent spinal degenerative disorders and imposes places heavy medical economic burdens on individuals society. Mechanical overloading applied to intervertebral (IVD) has been widely recognized as an important cause IVDD. overloading-induced chondrocyte ferroptosis was reported, but potential association between mechanical remains be illustrated in nucleus pulposus (NP) cells. In this study, we discovered that excessive loading induced endoplasmic reticulum (ER) stress, which were detected by mitochondria associated markers, increasing intracellular free Ca 2+ level through Piezo1 ion channel localized plasma membrane ER NP Besides, proposed elevation activation stress are positive feedback processes promote each other, consistent with results coccygeal discs aged Piezo1-CKO mice significantly lower than WT mice. Then, confirmed selenium supplementation decreased mitigating upregulating Selenoprotein K (SelK) expression. caused impaired production function Glutathione peroxidase 4 (GPX4) due calcium overload could improved Se-GPX4 axis Se-SelK vivo vitro, eventually presenting stabilization extracellular matrix (ECM). Our findings reveal role IVDD, promotes significance attenuate thus alleviates might provide insights into therapeutic interventions for

Lead (Pb), a heavy metal environmental pollutant, poses threat to the health of humans and birds. Inflammation is one most common pathological phenomena in case illness poisoning. However, underlying mechanisms inflammation remain unclear. The cerebellum thalamus are important parts nervous system. To date, there have been no reports Pb inducing animal cerebellums or thalami. Selenium (Se) can relieve Therefore, we aimed explore mechanism by which Se alleviates toxicity thalami chickens establishing chicken or/and treatment model. Our results demonstrated that exposure caused inflammatory damage thalami, evidenced characteristics inflammation, decrease anti-inflammatory factors (interleukin (IL)-2 interferon-γ (INF-γ)), increase pro-inflammatory (IL-4, IL-6, IL-12β, IL-17, nitric oxide (NO)). Moreover, found IL-2/IL-17–NO pathway took part Pb-caused injury. above findings were reversed supplementation dietary Se, meaning relieved via pathway. In addition, an up-regulated oxidative index malondialdehyde (MDA) two down-regulated antioxidant indices (glutathione (GSH) total capacity (TAC)) recorded after received stimulation, indicating excess oxidant/antioxidant imbalance stress, stress mediated GSH–IL-2 axis. Interestingly, inhibited four glutathione peroxidase (GPx) family members (GPx1, GPx2, GPx3, GPx4), three deiodinase (Dio) (Dio1, Dio2, Dio3), fifteen other selenoproteins (selenophosphate synthetase 2 (SPS2), selenoprotein (Sel)H, SelI, SelK, SelM, SelO, SelP1, SelPb, SelS, SelT, SelU, (Sep)n1, Sepw1, Sepx1, Sep15), suggesting reduced resulted involving SPS2–GPx1–GSH supplementation, as expected, changes mentioned above, improved mitigated chickens. For first time, discovered SPS2–GPx1–GSH–IL-2/IL-17–NO involved complex conclusion, this study Pb-induced This offers novel insights into pollutant-caused poisoning provides theoretical basis for detoxification effect against toxic pollutants.

Microplastics (MPs) generally refer to the plastic fragments or particles smaller than 5 mm in diameter, which are closely concerned due their widespread presence environment. Recent studies have shown that MPs a serious threat on reproductive health of organisms. Pigs often selected as model animals because high similarity human tissues and organs. However, there no reports effects mechanisms exposure swine germ cells. In present study, we established testis (ST) cell models exposed 250, 500, 1000 μg/ml polystyrene microplastics (PS-MPs, 1-10 μm), respectively. The findings revealed PS-MPs reduced viability dose-dependently. Acridine orange/ethidium bromide staining flow cytometry results indicated occurrence apoptosis necrosis ST cells under exposure, expression changes relevant marker genes (B-cell lymphoma-2, Bcl-2 Associated X, Caspase-3, Caspase-9, Receptor-interacting protein kinase 1, 3, Mixed lineage domain-like, Caspase-8) were clarified via quantitative real-time PCR western blot. Further mechanistic found treatment induced excessive intracellular reactive oxygen species (ROS) production, promoted phosphorylation mitogen-activated (MAPK) pathway-related (P38, c-Jun N-terminal kinase, extracellular regulated kinases) activated downstream gene hypoxia-inducible factor (HIF1α). conclusion, our study suggests causes necroptosis ROS/MAPK/HIF1α signaling pathway.
