Activating transcription factor 6 (ATF6), an important signaling molecule in unfolded protein response (UPR), plays a role the pathogenesis of several diseases, including diseases such as congenital retinal disease, liver fibrosis and ankylosing spondylitis. After endoplasmic reticulum stress (ERS), ATF6 is activated after separation from binding immunoglobulin (GRP78/BiP) (ER) transported to Golgi apparatus be hydrolyzed by site 1 2 proteases into fragments, which localize nucleus regulate expression ERS-related genes. In these ERS leads activation UPR, ultimately lead occurrence development regulating physiological state cells through pathway. Here, we discuss evidence for pathogenic importance different preclinical results.

Abstract Alzheimer’s disease (AD) is a multifactorial, rapidly progressing neurodegenerative disorder. As the exact cause of still unclear, drug development very challenging. This review encompasses commonly used AD models involving various chemicals, heavy metals and endogenous substances induced transgenic models. It also provides insight into reliable emerging that may overcome shortcomings associated with available Chemicals like streptozotocin, scopolamine, colchicine okadaic acid render animal susceptible to neuroinflammation oxidative stress neurodegeneration along amyloid-β deposition tau hyperphosphorylation. Similarly, acrolein 1–42 are efficient in inducing major pathologies AD. Heavy aluminum fluoride mixture these have been reported induce neurotoxicity therefore as for Transgenic developed result knock-in or knock-out certain genes including PDAPP, APP23, Tg2576, APP/PS1, 3 × Tg 5 FAD incorporated this study. Further, advanced pathomimetic provided particular interest here which will add on current knowledge aid process deepen our understanding related pathogenesis. Alzheimer's disease (AD) is a neurodegenerative that causes memory loss and progressive permanent deterioration of cognitive function. The most challenging issue in combating AD its complicated pathogenesis, which includes the deposition amyloid β (Aβ) plaques, intracellular hyperphosphorylated tau protein, neurofibrillary tangles (NFT), etc. Despite rapid advancements mechanistic research drug development for AD, currently developed drugs only improve ability temporarily relieve symptoms but cannot prevent AD. Moreover, blood–brain barrier (BBB) creates huge to delivery brain. Therefore, effective diagnostic tools treatments are urgently needed. In recent years, nanomedicine has provided opportunities overcome challenges limitations associated with traditional diagnostics or treatments. Various types nanoparticles (NPs) play an essential role diagnosis treatment acting as carriers targeting bioavailability across/bypass BBB directly on lesions. This review categorizes different NPs summarizes their applications It also discusses clinical explores latest developments prospects

Investigating amyloid-β (Aβ) peptides in solution is essential during the initial stages of developing lead compounds that can influence Aβ fibrillation while peptide still a soluble state. The tendency Aβ(1-42) to misfold solution, correlated aetiology Alzheimer's disease (AD), one main hindrances characterising its aggregation kinetics cell-mimetic environment. Moreover, triggers unfolded protein response (UPR) endoplasmic reticulum (ER), leading cellular dysfunction and multiple cell death modalities, exacerbated by reactive oxygen species (ROS), which damage components trigger inflammation. Antioxidants like curcumin, derivative Curcuma longa, help mitigate ER stress scavenging ROS enhancing antioxidant enzymes. Furthermore, evidence literature highlights effect curcumin on secondary structure Aβ(1-42). This explorative study investigates conformational behaviour presence six derivatives using circular dichroism (CD) explore their interactions with lipid bilayers, potentially preventing aggregate formation. results suggest synthetic tetrahydrocurcumin (THC) interacts amyloid all systems presented, cyclocurcumin (CYC) bisdemethoxycurcumin (BMDC) only interact when less stable conformation.
