Extracellular matrices (ECMs) are highly specialized and dynamic three-dimensional (3D) scaffolds into which cells reside in tissues. ECM is composed of a variety fibrillar components, such as collagens, fibronectin, elastin, non-fibrillar molecules proteoglycans, hyaluronan, glycoproteins including matricellular proteins. These macromolecular components interconnected forming complex networks that actively communicate with through binding to cell surface receptors and/or matrix effectors. ECMs exert diverse roles, either providing tissues structural integrity mechanical properties essential for tissue functions or regulating phenotype maintain homeostasis. molecular composition structure vary among tissues, markedly modified during normal repair well the progression various diseases. Abstract Fibrotic skin disease represents a major global healthcare burden, characterized by fibroblast hyperproliferation and excessive accumulation of extracellular matrix. Fibroblasts are found to be heterogeneous in multiple fibrotic diseases, but heterogeneity diseases is not well characterized. In this study, we explore keloid, paradigm using single-cell RNA-seq. Our results indicate that keloid fibroblasts can divided into 4 subpopulations: secretory-papillary, secretory-reticular, mesenchymal pro-inflammatory. Interestingly, the percentage subpopulation significantly increased compared normal scar. Functional studies crucial for collagen overexpression keloid. Increased also another disease, scleroderma, suggesting broad mechanism fibrosis. These findings will help us better understand pathogenesis, provide potential targets therapies.
