Oral gene therapy has emerged as a potential optimal treatment for ulcerative colitis (UC). Nucleic acid drugs possessing versatility can not only inhibit inflammation but realize colon mucosal healing, fulfilling the clinical objective of UC therapy. However, effective accumulation and distribution oral nucleic in remain considerable challenge. Furthermore, current delivery systems pay more attention to colon, while which plays key role treatment, never catches researchers. Here, we used miR-320 model drug develop kind multistage-responsive nanocomplexes (MSNs) based on polymeric nanocapsules alginate. MSNs possess pH responsiveness stomach, enzyme colonic lumen, redox cytoplasm. In vivo imaging results showed that reach within 2 h effectively release lumen. The further deliver submucosal layer even muscular layer. Moreover, decreased activity myeloperoxidase proinflammatory cytokines exhibited anti-inflammatory by inhibiting phosphorylation IκBα AKT, reducing enhancing repair. Therefore, successfully alleviate improving promoting translational application UC.

Cardiovascular diseases (CVDs) caused by arteriosclerosis are the leading cause of death and disability worldwide. In late stages atherosclerosis, atherosclerotic plaque gradually expands in blood vessels, resulting vascular stenosis. When unstable ruptures falls off, it blocks vessel causing thrombosis, to strokes, myocardial infarctions, a series other serious that endanger people's lives. Therefore, regulating stability is main means used address high mortality associated with CVDs. The progression complex integration cell apoptosis, lipid metabolism disorders, inflammatory infiltration, smooth muscle migration, neovascular infiltration. More recently, emerging evidence has demonstrated non-coding RNAs (ncRNAs) play significant role pathophysiological process formation affecting biological functions vasculature its cells. purpose this paper comprehensively review regulatory mechanisms involved susceptibility rupture, discuss limitations current approaches treat instability, highlight potential clinical value ncRNAs as novel diagnostic biomarkers therapeutic strategies improve reduce risk major cardiovascular events.

Ginsenoside Rg2 (Rg2) has a variety of pharmacological activities and provides benefits during inflammation, cancer, other diseases. However, there are no reports about the relationship between atherosclerosis. We used 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) to detect cell viability in vascular smooth muscle cells (VSMCs) human umbilical vein endothelial (HUVECs). The expression inflammatory factors HUVECs phenotypic transformation-related marker VSMCs were detected at mRNA levels. Western blot method was inflammation pathways transformation protein rat carotid balloon injury model performed explore effect on vivo.Rg2 decreased induced by lipopolysaccharide HUVECs-without affecting viability. These events depend blocking regulation NF-κB p-ERK signaling pathway. In VSMCs, can inhibit proliferation, migration, platelet derived growth factor-BB (PDGF-BB)-which may contribute its anti-atherosclerotic role. rats with injury, reduce intimal proliferation after regulate pathway response, also suppress VSMCs.These results suggest that exert cellular level animal level, which more sufficient basis for ginseng as functional dietary regulator.

Long noncoding RNAs (lncRNAs) are important regulators of various cellular functions. Recent studies have shown that a novel lncRNA termed Punisher is highly expressed in cardiovascular progenitors and has potential role diseases. However, its role, especially molecular mechanism, unclear. In our present study, we observed was obviously downregulated atherosclerotic plaques. Further research proved it can suppress the apoptosis VSMCs potentially contributing to progression atherosclerosis. Intriguingly, revealed regulate mitochondria fission as well mitochondrial functions induced by hydrogen peroxide (H2O2) VSMCs. Mechanistically, further serve ceRNA which directly binds miR-664a-5p consequently regulates target OPA1, finally contributes biological function Particularly, overexpression distinctly suppressed neointima formation VSMC vivo. Encouragingly, these results were accordance with findings obtained clinical evaluation patients Our data provides significant relationship among homeostasis, apoptosis, And could targets diagnosis treatment

Global warming is one of the most common environmental challenges faced by cold-water fish farming. Intestinal barrier function, gut microbiota, and microbial metabolites are significantly altered under heat stress, posing serious obstacles to healthy artificial culture rainbow trout. However, molecular mechanisms underlying intestinal injury in trout stress remain unclear. In present study, optimal growth temperature for (16 °C) was used control group, maximum tolerated (24 which subjected 21 days. The mechanism explored combining animal histology, 16S rRNA gene amplicon sequencing, ultra-high performance liquid chromatography-mass spectrometry, transcriptome sequencing. results showed that antioxidant capacity enhanced levels stress-related hormones were increased, relative expression genes related proteins indicating model successfully established. Secondly, tract inflammatory pathological characteristics with increased permeability, activation factor signaling pathway, genes, suggesting function impaired. Thirdly, caused an imbalance commensal microbiota changes trout, participated response mainly affecting lipid metabolism amino acid metabolism. Finally, promoted activating peroxisome proliferator-activated receptor-α pathway. These not only expand understanding physiology regulation mechanisms, but also provide a scientific basis reduction production costs.

Background: Diuretic resistance (DR) is characterized by insufficient fluid and sodium excretion enhancement despite maximum loop diuretic doses, indicating a phenotype of refractory heart failure (HF). Recently, metabolomics has emerged as crucial tool for diagnosing understanding the pathogenesis various diseases. This study aimed to differentiate diuretic-resistant patients from non-resistant HF identify biomarkers linked emergence DR. Methods: Serum samples patients, both with without DR, were subjected non-targeted metabolomic analysis using liquid chromatography-tandem mass spectrometry. Metabolite variations between groups identified principal component orthogonal partial least-square discriminant analysis. Metabolic pathways assessed through Kyoto Encyclopedia Genes Genomes database enrichment analysis, potential determined receiver operating characteristic curves (ROCs). Results: In total, 192 metabolites exhibited significant differences across two sample groups. Among these, up-regulation was observed in 164 metabolites, while 28 down-regulated. A total involving neuroactive ligand-receptor interaction amino acid biosynthesis affected. The top five ROC DR hydroxykynurenine, perillic acid, adrenic 5-acetamidovalerate, adipic acid. Conclusions: Significant metabolite profiles non-diuretic-resistant among HF. differentially expressed endogenous metabolic primary implicated noted energy, nucleotide metabolism.
