Abstract African swine fever (ASF) is an acute infectious haemorrhagic of pigs caused by virus (ASFV). Aloe-emodin (Ae) active ingredient Chinese herbs with antiviral, anticancer, and anti-inflammatory effects. We investigated the antiviral activity mechanism action Ae against ASFV using Real-time quantitative PCR (qPCR), western blotting, indirect immunofluorescence assays. significantly inhibited replication. Furthermore, transcriptomic analysis revealed that infection activated NF-κB signaling pathway in early stage apoptosis late stage. downregulated expression levels MyD88, phosphor-NF-κB p65, pIκB proteins as well mRNA IL-1β IL-8 porcine alveolar macrophages (PAMs) infected ASFV, thereby inhibiting activation induced ASFV. Flow cytometry blot increased percentage ASFV-induced apoptotic cells. Additionally, promoted upregulating cleaved-caspase3 Bax downregulating Bcl-2 proteins. This suggests promotes pathway, resulting inhibition These findings have further improved therapeutic reserves for prevention treatment ASF.

Abdominal obesity-related metabolic syndrome (MetS) has emerged as a significant global public health issue that affects human health. Flavonoids, such quercetin, have been reported to exert obvious anti-obesity and lipid-lowering effects in both humans animal models. However, the precise underlying mechanism remains elusive. In this study, we investigated potential roles of gut microbiota-bile acids (BAs) interactions quercetin-induced benefits. Oral administration quercetin significantly enhanced energy metabolism through activating thermogenesis brown adipose tissues (BAT) browning white (WAT), thus mitigating dysfunctions an abdominal MetS mouse model. Further mechanistic studies demonstrated treatment substantially promoted generation non-12α-hydroxylated BAs (non-12OH BAs), particularly ursodeoxycholic acid (UDCA) lithocholic (LCA), serum via regulating overall structure microbiota enriching Lactobacillus. High level non-12OH bind Takeda G protein-coupled receptor 5 (TGR5) on adipocytes stimulate thermogenesis. Remarkably, fecal transplantation (FMT) from quercetin-treated mice replicated expenditure, which suggested reshape concomitant regulation were responsible for benefits Our findings not only highlighted critical role microbiota-BAs crosstalk mediating but also enriched pharmacological mechanisms ameliorating MetS-related diseases.

Obesity is a global health concern with management strategies encompassing bariatric surgery and anti-obesity drugs; however, concerns regarding complexities side effects persist, driving research for more effective, low-risk strategies. The promotion of white adipose tissue (WAT) browning has emerged as promising approach. Moreover, alisol B 23-acetate (AB23A) demonstrated efficacy in addressing metabolic disorders, suggesting its potential therapeutic agent obesity management. Therefore, this study, we aimed to investigate the AB23A mitigating by regulating phenotypes lipid distribution mice fed high-fat diet (HFD). An mouse model was established administration an HFD. Glucose insulin metabolism were assessed via glucose tolerance tests. Adipocyte size determined using hematoxylin eosin staining. expression markers WAT evaluated Western blotting quantitative real-time polymerase chain reaction.
