Abstract Ferroptosis represents an emerging, iron-dependent form of cell death driven by lipid peroxidation. In recent years, it has garnered significant attention in the realm cancer immunotherapy, particularly studies involving immune checkpoint inhibitors. This not only enhances our comprehension tumor microenvironment but is also considered a promising therapeutic strategy to address resistance, investigate activation mechanisms, and facilitate development vaccines. The combination immunotherapy with ferroptosis provides innovative targets fresh perspectives for advancing treatment. Nevertheless, cells appear possess wider array evasion strategies compared CD8 + T cells, which have been conclusively shown be more vulnerable ferroptosis. Furthermore, TME can create favorable environment survival invasion. Under this premise, both inducing inhibiting will impact antitumor immunity some extent, even make final result run counter purpose. paper systematically elucidates dual-edged sword role process briefly outlining complexity within TME. It explores potential side effects associated ferroptosis-inducing therapies critically considers combined application ferroptosis-based ICIs. highlights current challenges faced approach points out future directions development.

Engineered biomaterials are materials specifically designed to interact with biological systems for biomedical applications. This paper offers the comprehensive analysis of global clinical trial trends involving such materials. We surveyed 834 studies in ClinicalTrials.gov database and explored biomaterial types, their initiation points, durations trials. Predominantly, synthetic natural polymers, particularly silicone collagen, used. Trials, focusing on ophthalmology, dentistry, vascular medicine, primarily conducted United States, Canada, Italy. These trials encompass a broad demographic, enrolled fewer than 100 participants. The study duration varied ranging from 0.5 4.5 years. mainly bioresorbable or bioinert, integration cells remaining an underexplored area. Our findings shed light current practices future potentials engineered research, offering insights advancing this dynamic field globally.

Hepatocellular carcinoma (HCC) is one of the most prevalent cancers worldwide and accounts for more than 90% primary liver cancer. The advent immune checkpoint inhibitor (ICI)-related therapies combined with angiogenesis inhibition has revolutionized treatment HCC in late-stage unresectable HCC, as ICIs alone were disappointing treating HCC. Immune checkpoint blockade (ICB) has become a promising strategy in treating advanced cancers, providing significant survival benefits for patients with various cancer types. However, among the vast population of patients, only small fraction are able to respond and derive from ICB therapy. Numerous factors contribute diminished efficacy ICB, complex tumor microenvironment (TME) playing an important role. Therefore, comprehensively understanding intricate composition TME is critical elucidating mechanisms that underlie distinct responses patients. Single-cell RNA sequencing (scRNA-seq) novel technique reveals gene expression profiles individual cells, facilitating investigation heterogeneity at high resolution identification key cell subsets participating response ICB. This review emphasizes importance scRNA-seq studying summarizes recent findings discovery biomarkers predict potential therapeutic targets immunotherapy. These suggest future directions clinical implementation immunotherapy, further advancements precision medicine.
