Flavonoids are naturally occurring dietary phytochemicals with significant antioxidant effects aside from several health benefits. People often consume them in combination other food components. Compiling data establishes a link between bioactive flavonoids and prevention of diseases animal models, including cardiovascular diseases, diabetes, gut dysbiosis, metabolic dysfunction-associated steatotic liver disease (MASLD). However, numerous clinical studies have demonstrated the ineffectiveness contradicting rodent thereby challenging validity using as supplements. This review provides perspective to emphasize effective roles well summarize their specific mechanisms animals briefly. First, this offers an in-depth elucidation processes within human, encompassing small, large intestine, liver. Furthermore, comprehensive overview various functions gastrointestinal tract, hindering breakdown assimilation macronutrients, such polysaccharides lipids, regulating hormone secretion inhibition mineral iron absorption. In unabsorbed major portion interact flora leading biotransformation. Once absorbed circulated bloodstream, or metabolites exert beneficial systemic effects. Lastly, we examine protective disorders, endothelial dysfunction, MASLD, disease, obesity, hyperlipidemia, insulin resistance. conclusion, outlines safety future prospects field health, especially syndrome (MetS).

ABSTRACT Hydroxytyrosol (HT) consumption is associated with healthy outcomes. Specifically, its ability to modulate microRNA (miRNA) expression has been described. However, whether HT influences the secretion of extracellular vesicles (EVs) and their transported miRNAs remains unknown. Here, following administration, we explored potential changes in EV miRNA cargo during postprandial state after 1 week daily intake. Twelve participants received, first, a single 25 mg dose (postprandial study) then daily, for one (sustained study). Plasma circulating plasma‐isolated EVs were analyzed derived metabolites using UPLC‐qTOF‐MS by RNA‐seq. induced 1‐h post‐ingestion 1‐week administration increased fasting reduced oxidized LDL levels. RNA‐seq analysis identified 55 modulated (30 upregulated downregulated) related response oxygen level gland development, supplementation. This pilot study sheds light on effects modulation exosomal humans, which scarcely studied date. Further research warranted elucidate molecular mechanisms underlying observed effects.

Background: Cellular senescence is a biological process with dual role in organismal health. While transient supports tissue repair and acts as tumor-suppressive mechanism, the chronic accumulation of senescent cells contributes to aging progression age-related diseases. Senotherapeutics, including senolytics, which selectively eliminate cells, senomorphics, modulate senescence-associated secretory phenotype (SASP), have emerged promising strategies for managing pathologies. Among these, polyphenols, diverse group plant-derived bioactive compounds, gained attention their potential cellular senescence. Methods: This review synthesizes evidence from vitro, vivo, clinical studies on senolytic senomorphic activities resveratrol, kaempferol, apigenin, fisetin. The analysis focuses molecular mechanisms action impact fundamental aging-related pathways. Results: Polyphenols exhibit therapeutic versatility by activating SIRT1, inhibiting NF-κB, modulating autophagy. These compounds demonstrate role, promoting survival healthy while inducing apoptosis cells. Preclinical indicates capacity reduce SASP-associated inflammation, restore homeostasis, attenuate various models aging. Conclusions: represent class senotherapeutics mitigating diseases lifespan extension. Further research should focus validation long-term effects these paving way development agents geriatric medicine.
