Reproductive system diseases pose prominent threats to human physical and mental well-being. Besides being influenced by genetic material regulation changes in lifestyle, the occurrence of these is closely connected exposure harmful substances environment. Endocrine disrupting chemicals (EDCs), characterized hormone-like effects, have a wide range influences on reproductive system. EDCs are ubiquitous natural environment present industrial everyday products. Currently, thousands been reported exhibit endocrine this number likely increase as testing for potential has not consistently required, obtaining data limited, partly due long latency many diseases. The ability avoid EDCs, especially those artificially synthesized origin, increasingly challenging. While can be divided into persistent non-persistent depending their degree degradation, recent uptick research studies area, we chosen focus pertaining detrimental effects health several that widely encountered daily life over past six years, specifically bisphenol A (BPA), phthalates (PAEs), polychlorinated biphenyls (PCBs), parabens, pesticides, heavy metals, so on. By focusing impact hypothalamic-pituitary-gonadal (HPG) axis, which leads development diseases, review aims provide new insights molecular mechanisms EDCs’ damage encourage further in-depth clarify potentially EDC through various other mechanisms. Ultimately, it offers scientific basis enhance risk management, an endeavor significant societal importance safeguarding health.

Endometriosis is a complex disease and still many areas of uncertainty exist on the clinical management, from diagnosis to treatment. The modern diagnostic workup should include family history (including genetic epigenetic factors), presentation, an accurate gynecological examination imaging evaluation (transvaginal ultrasound and/or magnetic resonance) performed by expert practitioners. Laparoscopy must no longer be with sole purpose diagnosing endometriosis. Furthermore, endometriosis already suspected among adolescents young women severe dysmenorrhea, interfering daily activities not responding analgesic drugs. For patients without immediate desire pregnancy, hormonal drugs are first line choices for treating endometriosis-related pain, aiming reduce menstruation frequency even abolishing any bleeding. Progestins or continuous combined oral contraceptives used long term treatments prescribed as first-line In case failure intolerance, GnRH analogs more recently antagonists may proposed (with add-back therapy). Surgery remains important treatment option, but it at right time, single operation, preferentially in referral centers dedicated multidisciplinary teams order avoid recurrences repetitive surgery. Assisted reproductive technology (ART) option infertility either approach, after surgical approach. Fertility preservation can discussed moment before performing surgery when indicated.

ABSTRACT Background Insulin resistance has been correlated with the genetic diversity within insulin‐like binding proteins genes. Moreover, insulin is one of key characteristics widespread reproductive endocrine condition known as polycystic ovarian syndrome (PCOS). Hence, this study aimed to determine association between IGFBP3 and IGF2BP2 gene variants PCOS risk. Methods A total 300 subjects (150 cases diagnosed based on Rotterdam ESHRE/ASRM consensus criteria 150 healthy subjects) were recruited in case–control cross‐sectional study. Tetra‐primer amplification refractory mutation system polymerase chain reaction (ARMS‐PCR) was used for genotyping rs11705701, whereas rs1470579 rs2854744 done employing PCR‐restriction fragment length polymorphism (PCR‐RFLP) technique. Results The CC AA+AC genotypes conferred an increased risk our population. Regarding rs2854744, observed under codominant homozygous (TT vs. GG) model by 2.54 fold. C allele T enhanced 1.97 1.46 folds, respectively. Haplotype analysis showed that rs11705701 haplotype a decreased (odds ratio = 0.53, 95% confidence interval 0.34–0.83, p 0.006). AC/GG/GT, AA/GA/GT, AC/GA/GG, AC/GA/GT genotype combinations rs1470579/rs11705701/rs2854744 associated disease. Conclusions susceptibility Southeastern Iranian Further investigation involving larger cohorts representing diverse ethnic backgrounds needed confirm current findings.

Endocrine-disrupting chemicals (EDCs) are exogenous compounds that interact with the estrogen receptor (ER), thereby disrupting estrogen-mediated signaling. In a previous study, we employed bioluminescence resonance energy transfer (BRET) system to assess ER dimerization for detecting EDCs. To further determine whether BRET assay could be used independently identify EDCs, investigated ER-EDC interactions before and after dimerization. Results from isothermal titration calorimetry (ITC) dynamic light scattering (DLS) revealed can mediated by Consequently, proved effective in clarifying its relevance EDC-induced signaling disruption. Additionally, examine transcriptional changes, performed chromatin immunoprecipitation sequencing (ChIP-seq), followed gene ontology (GO) analysis. These analyses demonstrated EDCs affect various pathways, including those involved antibody-dependent cytotoxicity, bone morphogenetic protein (BMP) cardiac induction, hepatocyte growth factor Overall, this study elucidates molecular mechanisms which influence findings highlight utility of BRET-based EDC detection contribute deeper understanding systemic effects on endocrine
