The COVID-19 pandemic has stimulated wastewater-based surveillance, allowing public health to track the epidemic by monitoring concentration of genetic fingerprints SARS-CoV-2 shed in wastewater infected individuals. Wastewater-based surveillance for is still its infancy. In particular, quantitative link between clinical cases observed through traditional and signals from viral concentrations developing hampers interpretation data actionable public-health decisions. We present a modelling framework that includes both transmission at population level fate RNA particles sewage system after faecal shedding persons population. Using our mechanistic representation combined clinical/wastewater system, we perform exploratory simulations quantify effect effectiveness, interventions vaccination on discordance signals. also apply model three Canadian cities provide wastewater-informed estimates actual prevalence, effective reproduction number incidence forecasts. find paired with this model, can complement supporting estimation key epidemiological metrics hence better triangulate state an using alternative source.

SUMMARY Wastewater-based surveillance (WBS) has undergone dramatic advancement in the context of coronavirus disease 2019 (COVID-19) pandemic. The power and potential this platform technology were rapidly realized when it became evident that not only did WBS-measured SARS-CoV-2 RNA correlate strongly with COVID-19 clinical within monitored populations but also, fact, functioned as a leading indicator. Teams from across globe innovated novel approaches by which wastewater could be collected diverse sewersheds ranging treatment plants (enabling community-level surveillance) to more granular locations including individual neighborhoods high-risk buildings such long-term care facilities (LTCF). Efficient processes enabled extraction concentration highly dilute matrix. Molecular genomic tools identify, quantify, characterize its various variants adapted programs applied these mixed environmental systems. Novel data-sharing allowed information mobilized made immediately available public health government decision-makers even public, enabling evidence-informed decision-making based on local dynamics. WBS since been recognized tool transformative potential, providing near-real-time cost-effective, objective, comprehensive, inclusive data changing prevalence measured analytes space time populations. However, consequence rapid innovation hundreds teams simultaneously, tremendous heterogeneity currently exists literature. This manuscript provides state-of-the-art review established details current work underway expanding scope other infectious targets.

Humanized mouse models and mouse-adapted SARS-CoV-2 virus are increasingly used to study COVID-19 pathogenesis, so it is important learn where the receptor ACE2 expressed. Here we mapped expression during postnatal development in adulthood. Pericytes CNS, heart, pancreas express strongly, as do perineurial adrenal fibroblasts, whereas endothelial cells not at any location analyzed. In a number of other organs, pericytes ACE2, including lung instead expressed bronchial epithelium alveolar type II cells. The onset organ specific: already birth, brain before, heart after day 10.5. Establishing vascular localization central correctly interpret data from modeling may shed light on cause complications.

The emergence of new variants SARS-CoV-2 associated with varying infectivity, pathogenicity, diagnosis, and effectiveness against treatments challenged the overall management COVID-19 pandemic. Wastewater surveillance (WWS), i.e., monitoring infections in communities through detecting viruses wastewater, was applied to track spread globally. However, there is a lack comprehensive understanding use WWS for variants. Here we systematically reviewed published articles reporting different wastewater by following PRISMA guidelines provided current state art this study area. A total 80 studies were found that reported until November 2022. Most these (66 out 80, 82.5%) conducted Europe North America, resource-rich countries. There high variation sampling strategy around world, composite (50/66 studies, 76%) as primary method In contrast, grab more common (8/14 57%) resource-limited Among detection methods, reverse transcriptase polymerase chain reaction (RT-PCR)-based sequencing quantitative RT-PCR commonly used wastewater. variants, B1.1.7 (Alpha) variant appeared earlier pandemic most (48/80 studies), followed B.1.617.2 (Delta), B.1.351 (Beta), P.1 (Gamma), others All same pattern clinical within timeline, demonstrating tracked all timely way when emerged. Wastewater-based epidemiology (WBE) has lately arised as a promising tool for monitoring and tracking viral pathogens in communities. In this study, we analysed WBE's role multi-pathogen surveillance strategy to detect the presence of several illness causative agents. Thus, an epidemiological study was conducted from October 2021 February 2023 estimate weekly levels Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), Syncytial virus (RSV), Influenza A (IAV) influent wastewater samples (n = 69). parallel, one-year (October 2022) performed assess pathogenic human enteric viruses. Besides, proposed fecal contamination indicators crAssphage Pepper mild mottle (PMMoV) also assessed, along with plaque counting somatic coliphages. Genetic material rotavirus (RV), astrovirus (HAStV), norovirus genogroup I (GI) GII found almost all samples, while hepatitis E viruses (HAV HEV) only tested positive 3.77 % 22.64 respectively. No seasonal patterns were overall viruses, although RVs had peak prevalence winter months. All SARS-CoV-2 RNA, mean concentration 5.43 log genome copies per liter (log GC/L). The circulating variants concern (VOCs) by both duplex RT-qPCR next generation sequencing (NGS). Both techniques reliably showed how dominant VOC transitioned Delta Omicron during two weeks Spain December 2021. RSV IAV peaked months concentrations 6.40 4.10 GC/L, Moreover, three selected respiratory strongly correlated reported clinical data when normalised physico-chemical parameters presented weaker correlations normalising sewage or coliphages titers. Finally, predictive models generated each virus, confirming high reliability on WBE early-warning system communities system. Overall, presents optimal reflecting circulation diseases trends within area, its value stands out due public health interest.
