Recently, the hallmarks of aging were updated to include dysbiosis, disabled macroautophagy, and chronic inflammation. In particular, low-grade inflammation during aging, without overt infection, is defined as "inflammaging," which associated with increased morbidity mortality in population. Emerging evidence suggests a bidirectional cyclical relationship between development age-related conditions, such cardiovascular diseases, neurodegeneration, cancer, frailty. How crosstalk other underlies biological mechanisms disease thus particular interest current geroscience research.

Age-associated osteoporosis (AAOP) poses a significant health burden, characterized by increased fracture risk due to declining bone mass and strength. Effective prevention early treatment strategies are crucial mitigate the disease burden associated healthcare costs. Current therapeutic approaches effectively target individual contributing factors AAOP. Nonetheless, management of AAOP is complicated multitude variables that affect its development. Main intrinsic extrinsic reviewed here, including mechanical unloading, nutrient deficiency, hormonal disbalance, disrupted metabolism, cognitive decline, inflammation circadian disruption. Furthermore, it discussed how these can be targeted for treatment. Although valuable as targets intervention, interconnectedness result in unique etiology every patient. Among these investigations, role antioxidants contrasting cognitive decline putting forward interesting promising results. In this review, we aim collect evidence that focused variety antioxidant-rich foods improving or stabilizing functions, memory, Alzheimer’s disease, most common disorder. Specifically, considered collected humans, either through longitudinal studies randomized, placebo-controlled ones, which evaluated performance, memory abilities, progression level neurodegeneration. Overall, despite between study protocols, cohorts participants involved, neuropsychological tests used, investigated antioxidants, there solid trend suggests properties may be helpful hampering older people. SUMMARY Health is strongly affected by aging and lifespan-modulating interventions, but the molecular mechanisms of mortality regulation remain unclear. Here, we conducted an RNA-seq analysis mice subjected to 20 compound treatments in Interventions Testing Program (ITP). By integrating it with data from over 4,000 rodent tissues representing responses genetic, pharmacological, dietary interventions established survival data, developed robust multi-tissue transcriptomic biomarkers mortality, capable quantifying change lifespan both short-lived long-lived models. These tools were further extended single-cell human demonstrating common across cell types species. Via a network analysis, identified annotated 26 co-regulated modules longevity tissues, interpretable module-specific clocks that capture aging- mortality-associated phenotypes functional components, including, among others, inflammatory response, mitochondrial function, lipid metabolism, extracellular matrix organization. captured characterized acceleration biological age induced progeria models chronic diseases rodents humans. They also revealed rejuvenation heterochronic parabiosis, early embryogenesis, cellular reprogramming, highlighting universal signatures shared age-related disease. included Cdkn1a Lgals3 , whose plasma levels demonstrated strong association all-cause disease incidence risk factors, such as obesity hypertension. Overall, this study uncovers hallmarks mammalian organs, types, species rejuvenation, exposing fundamental longevity.

Abstract Aging results from the gradual accumulation of molecular damage as a result cellular processes and is characterized by impaired functions, most notably an age-related decline in ATP production. However, causal relationship between homeostasis aging has not been established. In this study, we employed nucleotide transporter eukaryotic intracellular parasite to directly alter levels budding yeast cells exchange it with extracellular milieu. We found that depletion significantly shortens lifespan, whereas supplementation medium fully restores it. Analysis gene expression showed inhibition catabolic suggesting increased suppresses glucose metabolism. Our also leads lifespan extension. Overall, our study revealed direct impact on regulation lifespan. This work offers new insights into bioenergetic control positions energy metabolism promising target for longevity interventions.
