Atherosclerosis (AS) is a chronic inflammatory disease of the vascular wall with multiple causes. AS primary pathological basis cardiovascular and stroke. Moreover, carotid plaque rupture thrombus formation are main causes ischemic Therefore, understanding plaques may help improve prediction prevention cerebrovascular events. Endothelial cell dysfunction results in re‑endothelialization angiogenesis atherosclerotic plaques, thus promoting destabilization. The aim present study was to evaluate effect circular RNA (circRNA) molecules serum exosomes (serum‑Exos) from patients stable atherosclerosis (SA) unstable/vulnerable (UA). Specifically, circRNA on human umbilical vein endothelial (HUVEC) behavior mechanisms underlying destabilization were evaluated. Serum‑Exos isolated, then identified using transmission electron microscopy, nanoparticle tracking analysis western blotting. serum‑Exo‑circRNA expression profile SA or UA investigated array. relationship between circRNA‑006896 serum‑Exos biochemical parameters analyzed Spearman's correlation. In addition, HUVECs incubated for in vitro functional assays. demonstrated that circRNAs profiles significantly different, indicating potential role circRNA‑0006896 positively correlated triglyceride, low‑density lipoprotein cholesterol (LDL‑C) C‑reactive protein levels, negatively albumin levels UA. However, LDL‑C SA. Using bioinformatic analysis, competing endogenous (ceRNA) network selected regulatory roles serum‑Exos. Additionally, treated derived UA, upregulated. This accompanied by decreased microRNA‑1264 SOCS3, increased DNMT1 phosphorylated STAT3. HUVEC proliferation migration group, compared mock groups. finding indicates circRNA‑0006896‑miR-1264‑DNMT1 axis plays an important regulating cells. it suggests represent therapeutic target controlling JNK/STAT3 signaling HUVECs. Thus, this provide insight interventions against vulnerable AS.

Abstract Although androgen receptor (AR) can influence bladder cancer (BCa) initiation and progression, its impact on tumor immune escape remains unclear. Liraglutide has shown favourable effects on several cardiometabolic risk factors, beyond glucose control. MicroRNAs (miRNAs) regulate gene expression, resulting in post-transcriptional modifications of cell response and function. Specific miRNAs, including miRNA-27b, miRNA-130a, miRNA-210, play a role disease. We aimed to determine the effect liraglutide serum levels miRNA-130a miRNA-210. Twenty-five subjects with type-2 diabetes mellitus (T2DM), naïve incretin-based therapy, were treated (1.2 mg/day as an add-on metformin) for 4 months. miRNAs quantified using real-time polymerase chain reaction. After treatment, we found significant reductions fasting (from 9.8 ± 5.3 6.7 1.6 mmol/L, p = 0.0042), glycosylated haemoglobin (HbA1c) 8.1 0.8 6.6 1.0%, 0.0008), total cholesterol 5.0 1.0 4.0 0.7 0.0011), triglycerides 1.9 1.5 0.0104) low-density lipoprotein 2.9 1.2 2.2 0.6 0.0125), while miRNA-210a significantly increased (median (interquartile range, IQR) changes: 1.73 (7.12) (p 0.0401), 1.91 (3.64) 0.0401) 2.09 (11.0) 0.0486), respectively). Since changes independent all metabolic parameters investigated, seems exert direct epigenetic T2DM patients, regulating microRNAs involved maintenance endothelial homeostasis.
