Hepatic lipid droplets (LDs) and peroxynitrite (ONOO-) levels are closely related to nonalcoholic fatty liver disease (NAFLD). Additionally, some drug-induced injury (DILI) is often associated with ONOO-. Here, we constructed screened the quasi-LDs-targeted ONOO--responsive fluorescent probe MBDP-Py+ investigate interaction of NAFLD DILI. By monitoring upregulation ONOO- accumulation LDs, was more sensitive efficient than tissue staining serum markers detection in evaluating early toxicity diagnosing anticancer-DILI. More importantly, enhancement fluorescence signals demonstrated that different stages NAFLD, dominant element combined DILI mice models. As degree deepens, synergistic effect two will lead serious damage.

Intracellular peroxynitrite anions (ONOO-) and microenvironments (such as viscosity polarity) play an important role in maintaining redox homeostasis, regulating diffusion, transportation, signal transduction living cells. The abnormality of these factors is often closely related to various physiological/pathological processes. However, owing the lack suitable probes, simultaneous visualization ONOO-, viscosity, polarity ferroptosis cancer models has not been achieved. To meet urgent needs, we presented a multifunctional near-infrared (NIR) fluorescent probe, named MQA-P, for simultaneously detecting within mitochondria. probe exhibited remarkable turn-on response ONOO- with far-red emission about 645 nm was highly sensitive viscosity/polarity NIR channel λem > 704 nm. Facilitated by first time, revealed that erastin-induced accompanied significant upregulation increase (or decrease at same time. Moreover, concurrent use diagnosis successfully achieved only cell/tissue levels but also tumor mice models. Compared one factor, this detection multimarkers provides more reliable method/tool tracking ferroptosis-related pathological processes diagnosis, holding great potential preclinical research, medical imaging-guided surgery.
