Increasing evidence has demonstrated that amyloid-β peptide (Aβ), the hallmark of Alzheimer’s disease (AD), evokes oxidative and inflammatory cascades, which ultimately lead to death neurons. The purpose present study is demonstrate effect nobiletin, a representative compound citrus peel, in preventive therapeutic approaches against neuronal damage by exposure Aβ25–35. Nobiletin significantly ameliorated Aβ25–35-mediated cell restoring abnormal changes intracellular stress, cycle, nuclear morphology, activity apoptotic caspase. Regarding anti-inflammatory responses, nobiletin suppressed interleukin-1β, tumor necrosis factor-α, nitric oxide (NO), prostaglandin E2 production response Aβ stimulation. Moreover, inhibited Aβ-stimulated inducible NO synthase cyclooxygenase-2 expression, was attributed blockade factor-κB p65 phosphorylation its inhibitor, IκB-α. Interestingly, decreased expression c-Jun N-terminal kinase p38 without affecting extracellular signal-regulated 1/2 activation. Taken together, novel data implicate as potential candidate for prevention AD through inhibition apoptosis, inflammation.

Alzheimer’s disease (AD), which is characterized by the presence of amyloid-β (Aβ) plaques and neurofibrillary tangles, accompanied neurodegeneration, most common form age-related neurodegenerative disease. Parkinson’s (PD) second after AD, early prominent loss dopaminergic neurons in substantia nigra pars compacta. As currently available treatments are not able to significantly alter progression these diseases, successful therapeutic preventive interventions strongly needed. In course our survey substances from natural resources having anti-dementia neuroprotective activity, we found nobiletin, a polymethoxylated flavone peel Citrus depressa. Nobiletin improved cognitive deficits pathological features such as Aβ pathology, hyperphosphorylation tau, oxidative stress, animal models AD. addition, nobiletin motor PD models. These observations suggest that has potential become novel drug for treatment prevention diseases AD PD.

Neuroinflammation is involved in various neurological and neurodegenerative disorders which the activation of microglia one key factors. In this study, we examined anti-inflammatory effects flavonoids nobiletin (5,6,7,8,3′,4′-hexamethoxyflavone) eriodictyol (3′,4′,5,7-tetraxydroxyflavanone) on human cell line stimulated by either lipopolysaccharide (LPS), severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) full-length Spike protein (FL-Spike), or mycotoxin ochratoxin A (OTA). Human were preincubated with (10, 50, 100 µM) for h, following which, they 24 h. The inflammatory mediators interleukin-1 beta (IL-1β), chemokine (C-X-C motif) ligand 8 (CXCL8), IL-6, matrix metalloproteinase-9 (MMP-9) quantified culture supernatant enzyme-linked immunosorbent assay (ELISA). Both significantly inhibited LPS, FL-Spike, OTA-stimulated release IL-1β, CXCL8, MMP-9 at 50 µM, while, most cases, was also effective 10 pronounced reductions µM. These findings suggest that both are potent inhibitors pathogen-stimulated microglial mediators, highlighting their potential therapeutic application neuroinflammatory diseases, such as long COVID.
