Abstract Circadian clocks are ubiquitous timing mechanisms that generate approximately 24-h rhythms in cellular and bodily functions across nearly all living species. These internal clock systems enable organisms to anticipate respond daily changes their environment a timely manner, optimizing temporal physiology behaviors. Dysregulation of circadian by genetic environmental risk factors increases susceptibility multiple diseases, particularly cancers. A growing number studies have revealed dynamic crosstalk between cancer pathways, providing mechanistic insights into the therapeutic utility treatment. This review will discuss roles pathogenesis, highlighting recent advances chronotherapeutic approaches for improved

Cancer is the world's second most common cause of death, with colon cancer (CC) ranking third in prevalence and cancer-related fatalities. CC originates or rectum influenced by genetic mutations, lifestyle, diet. Although chemotherapy, especially 5-fluorouracil (5-FU), fundamental to treatment has improved patient survival, its benefits are often limited significant side effects emerging drug resistance. Recently, plant-based foods have drawn interest research for their natural bioactive compounds relatively low toxicity. Certain plant not only exhibit anti-cancer activity but may also boost effectiveness 5-FU while reducing adverse effects. Despite evidence suggesting a synergistic effect between 5-FU, this area remains fragmented. This review consolidates current on including fruits, vegetables, legumes, tea bioactives treatment. Besides, it explores mechanisms through which these enhance efficacy reduce effects, evaluates clinical application prospects, identifies challenges future directions. comprehensive analysis aims provide scientific foundation integrated CC, highlighting potential enhancing chemotherapy associated toxicities.

Drug resistance is the major obstacle of gemcitabine-based chemotherapy for treatment pancreatic ductal adenocarcinoma (PDAC). Many long non-coding RNAs (lncRNAs) are reported to play vital roles in cancer initiation and progression. Here, we report that lncRNA SLC7A11-AS1 involved gemcitabine PDAC. overexpressed PDAC tissues gemcitabine-resistant cell lines. Knockdown weakens stemness potentiates sensitivity resistant cells toward vitro vivo. promotes chemoresistance through reducing intracellular reactive oxygen species (ROS) by stabilizing nuclear factor erythroid-2-related 2 (NRF2), key regulator antioxidant defense. Mechanically, co-localized with β-TRCP1 nucleus. The exon 3 interacts F-box motif β-TRCP1, critical domain recruits SCFβ-TRCP E3 complex. This interaction prevents consequent ubiquitination proteasomal degradation NRF2 Our results demonstrate overexpression can scavenge ROS blocking SCFβ-TRCP-mediated NRF2, leading a low level ROS, which required maintenance stemness. These findings suggest as therapeutic target overcome treatment.

Neuroinflammation has been intensively demonstrated to be related various neurodegenerative diseases including Parkinson's disease (PD), amyotrophic lateral sclerosis (ALS), and Alzheimer's (AD). A natural polymethoxylated flavone, nobiletin (NOB) reported alleviate oxidative stress, insulin resistance, obesity. In this study, we evaluated the protection effects of NOB on neuroinflammation memory deficit. Three-month mice were administrated with by oral gavage every day for 6 weeks (100 mg/kg/day); subsequently injected intraperitoneally lipopolysaccharide (LPS) 7 days. Results behavioral tests revealed that dramatically ameliorated LPS-triggered deficit regarding synaptic dysfunctions neuronal loss. Also, suppressed microglial activation proinflammatory cytokine secretion, such as COX-2, IL-1β, TNF-α, iNOS. Similarly, upon LPS stimulation, pretreatment diminished secretion cytokines in BV-2 microglia cells exposure via modulating MAPKs, PI3K/AKT, NF-κB signaling pathways. addition, alleviated LPS-amplified redox imbalance, disturbance mitochondrial membrane potential (MMP), dampening expression protein respiration. The present study provides compelling evidence decreased LPS-stimulated impairment through maintaining cellular balance blocking transcriptional pathway, illustrating nutritional compound may serve a approach neuroinflammation-related diseases.
