People often keep engaging in behaviors that used to be successful the past but which are knowingly no longer effective current situation, so-called "action slips." Such action slips explained with stimulus-driven processes behavior is caused by a stimulus-response association and without information about outcome of behavior. This process contrasted goal-directed selected because it expected lead desired outcome. Failing act line changes taken as evidence for processes. Alcohol use disorder remains a substantial social, health, and economic problem drinking levels in women have been increasing recent years. Understanding whether how the underlying mechanisms that drive vary by sex is critical could provide novel, more targeted therapeutic treatments. Here, we examine results from our laboratories others which believe useful insights into similarities differences alcohol patterns across sexes. Findings for binge intake aversion-resistant, compulsion-like are considered, since both likely significant contributors to problems humans. We also describe studies regarding may underlie maladaptive drinking, with some focus on importance of nucleus accumbens (NAcb) core shell regions, several receptor types (dopamine, orexin, AMPA-type glutamate), possible contributions hormones. Finally, discuss stressors such as early life stress anxiety-like states interact contribute drinking. Together, these findings underscore relevance studying female male co-morbid conditions gain true clinically understanding addiction neuropsychiatric treatment.

Abstract This article reviews recent findings from the author’s laboratory that may provide new insights into how habits are made and broken. Habits extensively practiced behaviors automatically evoked by antecedent cues performed without their goal (or reinforcer) “in mind.” Goal-directed actions, in contrast, instrumental because is remembered valued. New results suggest actions transition to habit after extended practice when conditions encourage reduced attention behavior. Consistent with theories of learning, a behavior command less (and become habitual) as its reinforcer becomes well-predicted environment; learning prevented if presentation uncertain. Other not permanent, goal-direction can be restored several environmental manipulations, including exposure unexpected reinforcers or context change. more context-dependent than goal-directed are. Addiction is a complex disease that impacts millions of people around the world. Clinically, addiction formalized as substance use disorder (SUD), with three primary symptom categories: exaggerated use, social or lifestyle impairment, and risky use. Considerable efforts have been made to model features these criteria in non-human animal research subjects, for insight into underlying neurobiological mechanisms. Here we review evidence from rodent models SUD-inspired criteria, focusing on role striatal dopamine system. We identify distinct mesostriatal nigrostriatal circuit functions behavioral outcomes are relevant addictions SUDs. This work suggests essential not only positive SUDs, such elevated intake craving, but also impairments decision making underlie compulsive behavior, reduced sociality, risk taking. Understanding functional heterogeneity system related networks can offer this symptomatology may lead more targeted treatments.

Abstract Background Sex is an important factor in the progression and treatment of alcohol addiction, therapeutic approaches may have to be tailored potential sex differences. This highlights importance understanding differences behaviors that reflect key elements clinical such as continued use despite negative consequences (“compulsive use”). Studies experimental animals can help provide role plays influence these behaviors. Methods Large populations genetically heterogeneous male female Wistar rats were tested established model compulsive self-administration, operationalized responding contingent foot shock punishment. We also baseline (fixed ratio, unpunished) operant motivation self-administer (progressive ratio), temporal discounting for reward. In search predictors compulsivity, screened novelty-induced place preference, anxiety-like behavior, pain sensitivity corticosterone levels. The estrous cycle was monitored throughout study. Results Unpunished self-administration did not differ between males females when intake corrected body weight. Overall, showed higher levels alcohol. Compulsive response rates bimodal distributions but intermediate intensities used (0.2 0.25 mA); at intensities, uniformly suppressed both females. found less steep devalued by delaying its delivery. Males exhibited a stronger obtain under unpunished conditions, while baseline. Factor analysis underlying dimension related stress predicted compulsivity females, reward factor. find alcohol-related various stages cycle. Conclusions Our results suggest mechanisms promoting feature likely underscores considering biological variable preclinical research, has implications addiction.
