Triple-negative breast cancer (TNBC) has greater infiltration of M2-like macrophages (TAMs), which enhances cell invasion and leads to a poor prognosis. TNBC progression is mediated by both tumor cells the microenvironment (TME). Here we elucidate mechanism interaction between TAMs. In this study, confirmed that CD44v5 highly expressed in TNBC, drives metastasis promotes TAM polarization co-localizing with IL4Rα inhibiting its internalization degradation, thereby promoting activation STAT3/IL6 signaling axis. At same time, TAMs also facilitate secreting IL-4, IL-6, other cytokines, IL-4/IL-4R/STAT3/IL-6 axis plays role for responding Moreover, found above progress could be suppressed when domain was blocked.
