Abstract Circulating tumor cells (CTCs) is an established biomarker of cancer metastasis. The circulation dynamics CTCs are important for understanding the mechanisms underlying cell dissemination. Although studies have revealed that circadian rhythm may disrupt growth tumors, it generally unclear whether controls release CTCs. In clinical examinations, current in vitro methods detecting blood samples based on a fundamental assumption CTC counts peripheral do not change significantly over time, which being challenged by recent studies. Since practical to draw from patients repeatedly, feasible strategy investigate monitor them vivo detection methods. Fluorescence flow cytometry (IVFC) powerful optical technique able detect fluorescent circulating directly living animals noninvasive manner long period time. this study, we applied fluorescence IVFC noninvasively orthotopic mouse model human prostate cancer. We observed exhibited stochastic bursts progression. probability bursting activity was higher at early stages than late stages. longitudinally monitored 24-h period, and our results striking daily oscillations peaked onset night (active phase rodents), suggesting might be regulated rhythm.

Daylight stems solely from direct, scattered and reflected sunlight, undergoes dynamic changes in irradiance spectral power composition due to latitude, time of day, year the nature physical environment (reflections, buildings vegetation). Humans their ancestors evolved under these natural day/night cycles over millions years. Electric light, a relatively recent invention, interacts competes with light–dark cycle impact human biology. What are consequences living industrialised urban areas much less daylight more use electric throughout day (and at night), on general health quality life? In this workshop report, we have classified key gaps knowledge research into three main groups: (I) uncertainty as quantity needed for “optimal” physiological psychological functioning, (II) lack consensus practical measurement assessment methods tools monitoring real (day) light exposure across multiple scales, (III) insufficient integration exchange bases different disciplines. Crucial short long-term objectives fill proposed.

Chronic stress is well-known to cause physiological distress that leads body balance perturbations by altering signaling pathways in the neuroendocrine and sympathetic nervous systems. This increases allostatic load, which cost of fluctuations are required cope with psychological challenges as well changes physical environment. Recent studies have enriched our knowledge about role chronic disease development, especially carcinogenesis. Stress stimulates hypothalamic-pituitaryadrenal (HPA) axis system (SNS), resulting an abnormal release hormones. These activate elevate expression downstream oncogenes. occurs activation specific receptors promote numerous cancer biological processes, including proliferation, genomic instability, angiogenesis, metastasis, immune evasion metabolic disorders. Moreover, accumulating evidence has revealed β-adrenergic receptor (ADRB) antagonists target inhibitors exhibit remarkable anti-tumor effects. Circadian clocks are biological timing mechanisms that generate 24-h rhythms of physiology and behavior, exemplified by cycles sleep/wake, hormone release, metabolism. The adaptive value is evident when internal body daily environmental mismatched, such as in the case shift work jet lag or even mistimed eating, all which associated with physiological disruption disease. Studies animal human models have also unraveled an important role functional circadian modulating cellular organismal responses to cues (ex., food intake, exercise), pathological insults (e.g. virus parasite infections), medical interventions medication). The circadian system is composed of coupled endogenous oscillators that allow living beings, including humans, to anticipate and adapt daily changes in their environment. In mammals, clocks form a hierarchically organized network with ‘master clock’ located the suprachiasmatic nucleus hypothalamus, which ensures entrainment subsidiary environmental cycles. Robust rhythmicity body indispensable for temporally coordinating organ functions, disruption or misalignment rhythms caused instance by modern lifestyle strongly associated various widespread diseases. This review aims provide comprehensive overview our current knowledge about molecular architecture system‐level organization mammalian oscillators. Furthermore, we discuss regulatory roles peripheral cell physiology implication temporal coordination metabolism human health disease. Finally, summarize methods assessing humans.
