ABSTRACT Aging is considered the contributory accumulation of abruptions occurring through cell signaling cascades, which ultimately cause changes in physical functions, fate, and damage across all organ systems. DNA response (DDR) also occurs telomere shortening, tumor formation, mitochondrial dysfunction, so forth. Cellular aging cycle arrest, result extended DDR cascade networks via MDC1, 53BP1, H2AX, ATM, ARF, P53, P13‐Akt, BRAF, Sirtuins, NAD + , These persistent arrests initiated by other associated stress‐induced signals promote a permanent state arrest called senescence‐associated secretory phenotype (SASP). However, cellular gets accelerated with faulty repair systems, produced senescent cells further generate various promoting contributors to age‐related dysfunctional diseases including SASP. Any these factors contribute disease development. Therefore, this review explores anti‐aging targeting SASP regulation their detailed networks. In addition, it allows researchers identify targets therapeutic strategies based on identified nonidentified targets.

Abstract BACKGROUND Modern lifestyle has led to an increase in the age at conception. Advanced is one of critical risk factors for female-related infertility. It well known that maternal positively correlates with deterioration oocyte quality and chromosomal abnormalities oocytes embryos. The effect on endometrial function may be equally important factor influencing implantation rate, pregnancy overall female fertility. However, there are only a few published studies this topic, suggesting area been under-explored. Abstract Limited understanding exists regarding how aging impacts the cellular and molecular aspects of human ovary. This study combines single-cell RNA sequencing spatial transcriptomics to systematically characterize ovarian aging. Spatiotemporal signatures eight types cells during are observed. An analysis age-associated changes in gene expression reveals that DNA damage response may be a key biological pathway oocyte Three granulosa subtypes five theca stromal subtypes, as well their spatiotemporal aging, identified. FOXP1 emerges regulator declining with age inhibiting CDKN1A transcription. Silencing results premature insufficiency mice. These findings offer comprehensive variability aiding prioritization potential diagnostic biomarkers therapeutic strategies.

The ovarian microenvironment is critical for follicular development and oocyte maturation. Maternal conditions, including polycystic ovary syndrome (PCOS), endometriosis, aging, may compromise the microenvironment, development, quality. Chronic low-grade inflammation can induce oxidative stress tissue fibrosis in ovary. In PCOS, pro-inflammatory cytokine levels are often elevated fluids. women with obesity hyperandrogenemia insulin resistance chronic inflammation, thereby disrupting by increasing stress. endometrioma-derived iron overload stress, leading to ferroptosis fibrosis. inflammatory aging (inflammaging), senescent cells secrete senescence-associated secretory phenotype factors, causing Therefore, controlling would present a novel therapeutic strategy improving minimizing dysfunction.
