Platelet-rich fibrin (PRF), the coagulated plasma of fractionated blood, is widely used to support tissue regeneration in dentistry, and underlying cellular molecular mechanisms are increasingly being understood. Periodontal connective tissues steadily express CXCL8, a chemokine that attracts granulocytes lymphocytes, supporting homeostatic immunity. Even though PRF considered dampen inflammation, it should not be ruled out increases expression CXCL8 gingival fibroblasts. To test this hypothesis, we conducted bioassay where fibroblasts were exposed lysates respective serum. We show here and, lesser extent, serum increased by fibroblasts, as confirmed immunoassay. SB203580, inhibitor p38 mitogen-activated protein kinase, reduced expression. Consistently, weaker range, also caused phosphorylation Assuming rich source growth factors, TGF-β receptor type I kinase SB431542 decreased PRF-induced translation CXCL8. After tooth extraction, resorptive changes in extraction sockets and the adjacent alveolar ridge can affect subsequent replacement implantation. Several surgical concepts, including application of autologous blood concentrate platelet-rich fibrin (PRF), aim to reduce these changes. While PRF’s wound-healing pain-relieving effects are well-documented, its impact on bone regeneration is less clear due varying PRF protocols measurement methods for regeneration. Aim: The purpose of this study was to obtain data concerning growth factor release within liquid and solid platelet-rich fibrin (PRF) matrices estimate the amount potential interindividual variations as a basis for further preclinical clinical trials. Therefore, we aimed determine possible differences in factors between PRF. Materials Methods: Blood samples obtained from four subjects were processed both PRF using standard centrifugation protocol. Five (vascular endothelial factor, VEGF; epidermal EGF; platelet-derived factor-BB, PDGF-BB; transforming factor-β1, TGF-β1; matrix metallopeptidase 9, MMP-9) have been evaluated at six time points by ELISA over total observation period 10 days (1 h, 7 1 d, 2 d). Results: Growth could be measured all each point. Comparing matrices, no significant detected (p > 0.05). mean VEGF, TGFβ-1, PDGF-BB, MMP-9 raised peak point five (day 7) matrices. VEGF lower than PRF, whereas those PDGF-BB higher points. EGF had its already two after h (hour solid: = 180 pg/mL, SD 81; liquid: 218 64), declined rapidly until day 2, second slight on groups 182 189; 81 70). Conclusions: This with little variations. Chronic inflammation is a pathological process where cells of the mesenchymal lineage become major source inflammatory mediators. Platelet-rich fibrin (PRF) has been shown to possess potent anti-inflammatory activity in macrophages, but its impact on not investigated. Abstract In order to evaluate the therapeutic advantages of various autologous platelet concentrates (APC) as a single biomaterial during alveolar ridge preservation (ARP), systematic review with meta‐analyses was conducted. PubMed, EMBASE, Web Science, and Scopus were screened for randomized controlled trials (RCTs) that released prior 2024. The selected papers compared an APC either unassisted healing (blood clot) or another ARP (third molars not included). outcome parameters included bone dimension alterations, soft tissue healing, post‐op pain intensity. search yielded 35 (33 studies), one applying platelet‐rich plasma (PRP), six using rich in growth factors (PRGF), 28 leukocyte‐ fibrin (L‐PRF). These studies showed large heterogeneity (e.g., parameters, timing, surgical approach, inclusion criteria), which hindered drawing strong conclusions. most studies, however, PRP, PRGF, L‐PRF alone produced faster less post‐extraction pain, resorption, more socket fill, higher density when (spontaneous) healing. ultimate benefit appears be significantly influenced by approach. Limited literature exists comparing other biomaterials ARP, resulting inconclusive data. application is promising strategy improve hard reduce pain.

Abstract Introduction Alveolar ridge preservation (ARP) is a well‐defined treatment performed to reduce bone dimensional changes occurring during the healing of post‐extraction sockets allow for adequate implant placement. Leukocyte and platelet‐rich fibrin (L‐PRF) has been showing potentially promote tissue regeneration wound healing. Therefore, aim this study evaluate its efficacy ARP when applied fresh extraction sockets, in comparison with spontaneous Materials Methods Twenty‐seven patients hopeless non‐molar teeth were treated. After randomization, either filled L‐PRF or allowed heal spontaneously. CBCTs intraoral scans obtained immediately after at 4 months. Through superimposition images, horizontal width, height, buccal volume, contour measured, as well patient‐reported outcome measures (PROM's). Results The dimensions changed similarly both groups. Although less reduction occurred test group 1 mm from crest, differences not statistically significant ( p > 0.05). Application did prevent reductions contours, neither linear vertical aspect nor volumetric changes. There no between groups need placing implants. Patients reported similar outcomes terms bleeding, pain, inflammation, function weeks postoperatively. Conclusion minimized resorption tooth sites reduced Furthermore, use improve PROM's.
