Aging is characterized by a chronic low-grade sterile inflammation dubbed as inflammaging, which in part originates from accumulating cellular debris. These, acting danger signals with many intrinsic factors such cytokines, are sensed network of pattern recognition receptors and other cognate receptors, leading to the activation inflammasomes. Due inflammasome activity-dependent increase levels pro-inflammatory interleukins (IL-1β, IL-18), initiated, resulting tissue injury various organs, brain spinal cord included. Similarly, age-related diseases central nervous system (CNS), prominent moment, cells neurovascular unit occupy significant position. In this review, we discuss inflammatory changes normal aging summarize current knowledge on role inflammasomes contributing mechanisms common CNS diseases, namely Alzheimer’s disease, Parkinson’s amyotrophic lateral sclerosis stroke, all occur more frequently aging.

Inflammation is a major risk factor of morbidity and mortality in older adults. Although its precise etiology unknown, low-grade inflammation adults commonly associated with increased intestinal epithelial permeability (leaky gut) abnormal (dysbiotic) gut microbiota. The increasing population lack treatments to reduce aging-related microbiota dysbiosis, leaky gut, culminates rise comorbidities, constituting significant public health concern. Here, we demonstrate that human-origin probiotic cocktail containing 5 Lactobacillus Enterococcus strains isolated from healthy infant prevented high-fat diet–induced (HFD-induced) inflammation, metabolic dysfunctions, physical function decline mice. Probiotic-modulated primarily reduced by tight junctions, which turn inflammation. Mechanistically, probiotics modulated way increase bile salt hydrolase activity, taurine abundance the stimulated junctions suppressed leakiness. Furthermore, Caenorhabditis elegans, life span, adiposity enhanced function. results suggest such therapies could prevent or treat elderly.

Curcumin is a compound isolated from turmeric, plant known for its medicinal use. Recently, there growing interest in the medical community identifying novel, low-cost, safe molecules that may be used treatment of inflammatory and neoplastic diseases. An increasing amount evidence suggests curcumin represent an effective agent several skin conditions. We examined most relevant vitro vivo studies published to date regarding use inflammatory, neoplastic, infectious diseases, providing information on bioavailability safety profile. Moreover, we performed computational analysis about curcumin's interaction towards major enzymatic targets identified literature. Our results suggest well-tolerated, However, bypass limitations (low oral bioavailability, metabolism) essential order conduct larger clinical trials could confirm these observations. The possible combination with traditional drugs formulations novel delivery systems very promising field future applicative research.

Dietary odd-chain saturated fatty acids (OCFAs) are present in trace levels dairy fat and some fish plants. Higher circulating concentrations of OCFAs, pentadecanoic acid (C15:0) heptadecanoic (C17:0), associated with lower risks cardiometabolic diseases, higher dietary intake OCFAs is mortality. Population-wide OCFA levels, however, have been declining over recent years. Here, we show C15:0 as an active that attenuates inflammation, anemia, dyslipidemia, fibrosis vivo, potentially by binding to key metabolic regulators repairing mitochondrial function. This the first demonstration C15:0's direct role attenuating multiple comorbidities using relevant physiological mechanisms at established concentrations. Pairing our findings evidence (1) not readily made endogenously, (2) blood mortality a poorer state, (3) has demonstrated activities efficacy parallel health benefits humans, propose potential essential acid. Further studies needed evaluate impact decades reduced OCFA-containing foods contributors deficiencies susceptibilities chronic disease.
