Preservation and long-term storage of readily available cell-laden tissue-engineered products are major challenges in expanding their applications healthcare. In recent years, there has been increasing interest the development off-the-shelf using cryobioprinting approach. Here, bioinks incorporated with cryoprotective agents (CPAs) to allow fabrication cryopreservable tissue constructs. Although this method shown potential products, impact CPAs on viscoelastic behavior printability at cryo conditions remains unexplored. study, we have evaluated influence such as glycerol dimethyl sulfoxide (DMSO) rheological properties pre-crosslinked alginate for cryoprinting applications. DMSO-incorporated showed a reduction viscosity yield stress, while addition improved both due interactions calcium chloride used pre-crosslinking. Further, tube inversion experiments were performed identify suitable concentrations containing & CaCl2. Finally, based analysis cell recovery results, 10% was preservation constructs −80 °C viability cells within printed structures after recovery. Cell results indicate that bioink significantly compared without CPAs, confirming suitability developed combination fabricate on-demand

Biodiversity is defined as the presence of a variety living organisms on Earth that essential for human survival. However, anthropogenic activities are causing sixth mass extinction, threatening even our own species. For many animals, dwindling numbers becoming fragmented populations with low genetic diversity, long-term species viability. With extinction rates 1000-10,000 times greater than natural,

Abstract Background In vitro studies using nucleus pulposus (NP) cells are commonly used to investigate disc cell biology and pathogenesis, or aid in the development of new therapies. However, lab‐to‐lab variability jeopardizes much‐needed progress field. Here, an international group spine scientists collaborated standardize extraction expansion techniques for NP reduce variability, improve comparability between labs utilization funding resources. Methods The most applied methods extraction, expansion, re‐differentiation were identified a questionnaire research groups worldwide. from rat, rabbit, pig, dog, cow, human tissue experimentally assessed. Expansion media also investigated. Results Recommended protocols provided common species utilized culture. Conclusions This international, multilab multispecies study greater yield fewer gene expression changes by applying species‐specific pronase usage, 60–100 U/ml collagenase shorter durations. Cryoprotective and cytoprotective agents (Cytoprotective Agents) are fundamental components of the cryopreservation process. This review presents essentials process by examining its drawbacks role in protecting cell physiology. Natural cryoprotective agents, such as antifreeze proteins, sugars natural deep eutectic systems, have been compared with synthetic ones, addressing their mechanisms action efficacy protection. The final part this article focuses melatonin, a hormonal substance antioxidant properties, emerging agent for somatic cells gametes, including ovarian tissue, spermatozoa spermatogonial stem cells.
