Abstract Circular RNAs (circRNA) are a class of covalently closed single-stranded that have been implicated in cancer progression. Here we identify circNDUFB2 to be downregulated non-small cell lung (NSCLC) tissues, and negatively correlate with NSCLC malignant features. Elevated inhibits growth metastasis cells. Mechanistically, functions as scaffold enhance the interaction between TRIM25 IGF2BPs, positive regulator tumor progression metastasis. This TRIM25/circNDUFB2/IGF2BPs ternary complex facilitates ubiquitination degradation this effect enhanced by N 6 -methyladenosine (m A) modification . Moreover, is also recognized RIG-I activate RIG-I-MAVS signaling cascades recruit immune cells into microenvironment (TME). Our data thus provide evidences participates IGF2BPs activation anti-tumor immunity during via modulation both protein degradation, well cellular responses.

ABSTRACT Neuropeptides are a diverse class of neuronal signalling molecules that regulate physiological processes and behaviour in animals. However, determining the relationships evolutionary origins heterogeneous assemblage neuropeptides identified range phyla has presented huge challenge for comparative physiologists. Here, we review revolutionary insights into evolution neuropeptide have been obtained recently through analysis genome/transcriptome sequence data by ‘deorphanisation’ receptors. The at least 30 systems traced to common ancestor protostomes deuterostomes. Furthermore, two rounds genome duplication gave rise an expanded repertoire vertebrate lineage, enabling neofunctionalisation and/or subfunctionalisation, but with lineage-specific gene loss additional or duplications generating complex patterns phylogenetic distribution paralogous systems. Abstract Background The spider Trichonephila antipodiana (Araneidae), commonly known as the batik golden web spider, preys on arthropods with body sizes ranging from ∼2 mm in length to insects larger than itself (>20‒50 mm), indicating its polyphagy and strong dietary detoxification abilities. Although it has been reported that an ancient whole-genome duplication event occurred spiders, lack of a high-quality genome limited characterization this event. Results We present chromosome-level T. constructed basis PacBio Hi-C sequencing. assembled is 2.29 Gb size scaffold N50 172.89 Mb. scaffolding assigned 98.5% bases 13 pseudo-chromosomes, BUSCO completeness analysis revealed assembly included 94.8% complete arthropod universal single-copy orthologs (n = 1,066). Repetitive elements account for 59.21% genome. predicted 19,001 protein-coding genes, which 96.78% were supported by transcriptome-based evidence 96.32% matched protein records UniProt database. also shows substantial expansions several detoxification-associated gene families, including cytochrome P450 mono-oxygenases, carboxyl/cholinesterases, glutathione-S-transferases, ATP-binding cassette transporters, reflecting possible genomic polyphagy. Further architecture reveals event, based 2 lines evidence: (i) large-scale duplications inter-chromosome synteny (ii) duplicated clusters Hox genes. Conclusions represents valuable resource research provides insights into species’ adaptation environment.

Abstract Background Arthropods comprise the largest and most diverse phylum on Earth play vital roles in nearly every ecosystem. Their diversity stems part from variations a conserved body plan, resulting recorded adaptive changes genome. Dissection of genomic record sequence change enables broad questions regarding genome evolution to be addressed, even across hyper-diverse taxa within arthropods. Results Using 76 whole sequences representing 21 orders spanning more than 500 million years arthropod evolution, we document gene protein domain content provide temporal phylogenetic context for interpreting these innovations. We identify many novel families that arose early arthropods during diversification insects into modern orders. reveal unexpected variation patterns DNA methylation examples family coincident with appearance notable phenotypic physiological adaptations such as flight, metamorphosis, sociality, chemoperception.
