Liver cancer is one of the most common tumors with a high malignant degree in world. Its diagnosis and treatment are very difficult limited. More novel powerful DAT strategies urgently needed to break this situation. An increasing number studies have shown that microRNAs (miRNAs) could be used not only as biomarkers for prognosis hepatocellular carcinoma (HCC) but also important targets molecular targeted therapy. However, role miR-550a-5p HCC its specific mechanism remain unclear. Here we proposed verified hypothesis regulate progression was positively associated poor prognosis. Hepatocellular carcinoma (HCC), a leading cause of cancer-related deaths globally, poses significant challenges in early detection. Improved diagnostic accuracy can drastically influence patient outcomes, emphasizing the need for innovative, non-invasive biomarkers. This study utilized cohort 402 participants, including healthy controls, chronic hepatitis patients, and HCC patients from Bangladesh, to evaluate DNA methylation signatures peripheral blood mononuclear cells (PBMC). We performed targeted next-generation sequencing on selected genes previously identified assess their dynamics. The development M8 M4 scores was based these dynamics, using Receiver Operating Characteristic (ROC) analysis determine effectiveness detecting early-stage alongside existing markers such as epiLiver alpha-fetoprotein (AFP). Integration with AFP significantly enhances sensitivity HCC. Abstract The aim of this study is to explore the prognostic value vascular invasion (VI) in hepatocellular carcinoma (HCC) by searching for competing endogenous RNAs (ceRNA) network and constructing a new model HCC. differentially expressed genes (DEGs) between HCC normal tissues were identified from GEO TCGA. StarBase miRanda prediction tools applied construct circRNA-miRNA-mRNA network. DEGs with without VI also identified. Then, hub screened build risk score through method least absolute shrinkage selection operator. ability was assessed using Kaplan−Meier Cox regression analysis. In result, there 221 up-regulated 47 down-regulated circRNAs (DEcircRNAs) compared tissue. A circRNA-related ceRNA established, containing 11 DEcircRNAs, 12 DEmiRNAs, 161 DEmRNAs. Meanwhile, another DEG analysis revealed 625 123 VI, then protein–protein interaction (PPI) built based on 122 VI-related DEGs.
