Abstract: Inflammatory cytokines, interleukin-36 (IL-36), IL-37, IL-38 belong to IL-1 family. The IL-36 subfamily obtains pro- and anti-inflammatory effects on various immune responses. Cytokine has functions in immunity, the recently identified negatively associated with disease pathogenesis. To date, expression of IL-36, is reported dysregulated osteoarthritis (OA) rheumatoid arthritis (RA), may be markers for arthritis-related diseases. Interestingly, was different either OA patients or animal models, IL-36Ra synovium RA patients. Moreover, functional studies have demonstrated significant role these cytokines progress. These processes were related cells non-immune cells, where regulate downstream signalings then involve OA, development. In this review, we comprehensively discuss recent advancements development RA. We hope that targeting will become a potential treatment option future. Keywords: IL-38, osteoarthritis,

Abstract Developing effective antimicrobial biomaterials is a relevant and fast‐growing field in advanced healthcare materials. Several well‐known (e.g., traditional antibiotics, silver, copper etc.) newer nanostructured, chemical, biomimetic approaches have been researched developed recent years valuable knowledge has gained. However, associated infections (BAIs) remain largely unsolved problem breakthroughs this area are sparse. Hence, novel high risk potential gain needed to address the important challenge of BAIs. Antibiotic free that based on physical action promising, since they reduce antibiotic resistance tolerance. Here, selected examples reviewed such biomaterials, namely switchable, protein‐based, carbon‐based bioactive glass, considering microbiological aspects The review shows mainly powerful tools control microbial growth at interfaces. A robust adaptive immune response is essential for combatting pathogens. In the wrong context such as due to genetic and environmental factors, however, same mechanisms crucial self-preservation can lead a loss of self-tolerance. Resulting autoimmunity manifests in development host organ-specific or systemic autoimmune diseases, hallmarked by aberrant responses tissue damage. The prevalence diseases on rise, medical management which focuses primarily pharmacological immunosuppression that places patients at risk side effects, including opportunistic infections tumorigenesis. Biomaterial-based drug delivery systems confer many opportunities address challenges associated with conventional disease management. Hydrogels, particular, protect encapsulated cargo (drug cell therapeutics) from environment, afford their presentation controlled manner, be tailored respond conditions support treatment via multiplexed functionality. Moreover, localized affected sites these approaches has potential concentrate action site, reduce off-target exposure, enhance patient compliance reducing need frequent administration. Despite benefits disease, biomaterial-based focus largely downstream effects hypersensitivity have limited capacity eradicate disease. contrast, direct targeting reactions uniquely enables prophylaxis arrest progression mitigating basis autoimmunity. One promising approach induce self-antigen-specific tolerance, specifically subdues damaging autoreactivity while otherwise retaining normal responses. this review, we will discuss hydrogel-based those target head-on. As field continues advance, it expand range therapeutic choices people coping providing fresh prospects better clinical outcomes improved quality life.

Rheumatoid arthritis (RA) requires therapeutic approaches that alleviate symptoms and inhibit the progression of joint damage. Glucocorticoids (GCs) have been a cornerstone RA treatment, yet their use is often limited by side effects. Recent advancements suggest liposome-based delivery systems can improve GC biodistribution, minimizing toxicity. This study introduces an innovative tool for treatment using prednisone-encapsulated nonphospholipid liposomes (NPLs) in combination with hyaluronic acid (HA) hydrogel. Our methodology involved incorporating prednisone (PR) palmitic cholesterol to formulate stable NPLs thin-film hydration technique. The synthesized PR-NPLs, characterized mean size 150 nm, demonstrated uniform distribution higher drug encapsulation comparison conventional phospholipid liposomes. In vitro assays revealed PR-NPL markedly reduced inflammatory responses macrophages. Additionally, we successfully incorporated into HA hydrogel, employing photoinitiated cross-linking process. novel composite offered modulable PR release, governed degree hydrogel cross-linking. developed system presents promising advancement management, especially suited intraarticular injections. It potentially enables targeted, controlled release risk effects, signifying significant improvement over existing therapies.

Osteoarthritis and rheumatoid arthritis, though etiologically distinct, are both inflammatory joint diseases that cause progressive injury, chronic pain, loss of function. Therefore, long-term treatment with a focus on relieving symptoms is needed. At present, the primary for arthritis drug therapy, oral intravenous. Although significant progress has been achieved these methods in alleviating symptoms, certain prominent drawbacks such as substantial side effects limited absorption medications call an urgent need improved delivery methods. Injected hydrogels can be used system to deliver drugs cavity controlled manner continuously release them, thereby enhancing retention improve therapeutic effectiveness, which attributed desirable attributes low immunogenicity, good biodegradability biocompatibility.
