Background Liver cancer's severity has prompted interest in herbal remedies for hepatotoxicity and the disease. Acacia nilotica (Vachellia nilotica), valued its traditional medicinal properties, particularly aerial components, is being investigated hepatoprotective anti-cancer effects. However, research on A. bark extracts their mechanisms of action limited. Materials Methods The study aimed to determine Total Phenolic Content (TPC), Flavonoid (TFC), antioxidant properties extract using biochemical assays. Additionally, it examined protective effects normal Cells (LO2) toxicity against liver cancer cells (HepG2) cell culture techniques. Results showed significant (EC50-16.34), with a total phenolic content 159.98±9.91 mg GAE/DW flavonoid 16.93 QE/g dry weight. It exhibited moderate towards HepG2 at concentrations >100 μg/mL but was non-toxic LO2 cells. Moreover, prevented oxidative damage induced by H2O2. Conclusion These findings highlight therapeutic potential treatment, warranting further clinical investigation.
