Abstract Hyperuricemia, characterized by elevated levels of serum uric acid (SUA), is linked to a spectrum commodities such as gout, cardiovascular diseases, renal disorders, metabolic syndrome, and diabetes, etc. Significantly impairing the quality life for those affected, prevalence hyperuricemia an upward trend globally, especially in most developed countries. UA possesses multifaceted role, antioxidant, pro-oxidative, pro-inflammatory, nitric oxide modulating, anti-aging, immune effects, which are significant both physiological pathological contexts. The equilibrium circulating urate hinges on interplay between production excretion, delicate balance orchestrated transporter functions across various epithelial tissues cell types. While existing research has identified involvement numerous biological processes signaling pathways, precise mechanisms connecting disease etiology remain be fully elucidated. In addition, influence genetic susceptibilities environmental determinants calls detailed nuanced examination. This review compiles data from global epidemiological studies clinical practices, exploring foundations transporters depth. Furthermore, we uncover complex induced inflammation influences individuals with association its relative disease, offering foundation innovative therapeutic approaches advanced pharmacological strategies.

The human gastrointestinal tract is home to a complex microbial community that plays an important role in the general well-being of entire organism. gut microbiota generates variety metabolites and thereby regulates many biological processes, such as regulation immune system. In gut, bacteria are direct contact with host. major challenge here prevent unwanted inflammatory reactions on one hand other ensure system can be activated when pathogens invade. Here REDOX equilibrium utmost importance. This controlled by either directly or indirectly via bacterial-derived metabolites. A balanced microbiome sorts for stable balance, whereas dysbiosis destabilizes this equilibrium. An imbalanced status affects disrupting intracellular signaling promoting responses. we (i) focus most common reactive oxygen species (ROS) (ii) define transition from state oxidative stress. Further, (iii) describe ROS regulating Thereafter, (iv) examine influence homeostasis how shifts pro- anti-oxidative cellular conditions suppress promote responses inflammation.

The goal of this study was to examine the role sodium butyrate in preserving intestinal mucosal barrier and reducing hyperuricemia (HUA).First, we established a mouse model HUA via intraperitoneal injection potassium oxonate together with yeast-rich diet detect levels serum uric acid (UA) fecal short-chain fatty acids (SCFAs). Then, vitro, different concentrations UA (NaB) were used treat LS174T Caco2 cells. effects NaB on gut determined based expression MUC2, ZO-1, Occludin. Finally, C57BL/6 mice HUA, these administered 200 mg·kg-1·d-1 by gavage counter HUA. effect tract elucidated determining levels, inflammatory parameters, epithelial integrity, histological analysis. The data showed that content SCFAs decreased. Additionally, cells, reversed decrease Occludin, MUC2 protein caused high levels. Furthermore, decreased mice, both ABCG2 proteins increased level factors tissues mice. The damage. protected reduced level.

Resveratrol (RES) has been reported to prevent hyperuricemia (HUA); however, its effect on intestinal uric acid metabolism remains unclear. This study evaluated the impact of RES in mice with HUA induced by a high-fat diet (HFD). Moreover, we revealed underlying mechanism through metagenomics, fecal microbiota transplantation (FMT), and 16S ribosomal RNA analysis. We demonstrated that reduced serum acid, creatinine, urea nitrogen, urinary protein levels, improved glomerular atrophy, unclear renal tubule structure, fibrosis, inflammation. The results also showed increased degradation. significantly changed flora composition HFD-fed enriching beneficial bacteria degrade reducing harmful promote inflammation, improving microbial function via upregulation purine metabolism. FMT further is essential for HUA, Lactobacillus may play key role this process. present alleviates HFD-induced injury regulating gut acid.
