Abstract MicroRNAs (miRNAs) are a group of small non-coding RNAs that regulate gene expression by targeting mRNA. Moreover, it has been shown miRNAs changed in various diseases, such as cancers, autoimmune disease, infectious and neurodegenerative Diseases. The suppression miRNA function can be easily attained utilizing anti-miRNAs. In contrast, an enhancement achieved through the utilization modified mimetics. discovery appropriate carriers body become interesting subject for investigators. Exosomes (EXOs) therapeutic efficiency safety transferring different cellular biological components to recipient cell have attracted significant attention their capability carriers. Mesenchymal stem cells (MSCs) recognized generate wide range EXOs (MSC-EXOs), showing MSCs may effective EXO generation clinically measure compared other origins. MSC-EXOs widely investigated because immune attributes, tumor-homing flexible characteristics. this article, we summarized features MSC-EXOs, including production, purification, loading methods modification targeted delivery diseases. Rheumatoid arthritis (RA) is a chronic inflammatory joint disorder that causes systemic inflammation, autoimmunity, and abnormalities result in permanent disability. Exosomes are nanosized extracellular particles found mammals (40–100 nm). They transporter of lipids, proteins, genetic material involved mammalian cell–cell signaling, biological processes, cell signaling. have been identified as playing role rheumatoid arthritis-related inflammation (RA). Uniquely functioning vesicles (EVs) responsible for the transport autoantigens mediators between distant cells. In addition, paracrine factors, such exosomes, modulate immunomodulatory function mesenchymal stem cells (MSCs). addition to transporting information, exosomes convey miRNAs studied drug delivery vehicles. animal models, it has observed MSCs secrete EVs with properties, promising results this area. By understanding diversity exosomal contents their corresponding targets, may be possible diagnose autoimmune diseases. can employed diagnostic biomarkers immunological disorders. We here discuss most recent findings regarding diagnostic, prognostic, therapeutic potential these nanoparticles provide an overview evidence pertaining biology RA.

Abstract Extracellular vesicles (EVs) are nanosized with a lipid bilayer that secreted by cells and play critical role in cell‐to‐cell communication. Despite the promising reports regarding their diagnostic therapeutic potential, utilization of EVs clinical setting is limited due to insufficient information about cargo lack standardization isolation analysis methods. Considering protein cargos as key contributors potency, we conducted tandem mass tag (TMT) quantitative proteomics three subpopulations mesenchymal stem cell (MSC)‐derived obtained through different techniques: ultracentrifugation (UC), high‐speed centrifugation (HS), on sucrose cushion (SU). Subsequently, checked EV marker expression, size distribution, morphological characterization, followed bioinformatic analysis. The proteome results revealed these exhibit distinct molecular functional characteristics. choice method impacts isolated isolating EVs. Specifically, (HS) exhibited higher abundance ribosomal mitochondrial proteins. Functional apoptosis assays comparing mitochondria methods HS‐EVs, but not other EVs, induced early cancer cells. On hand, using (SU) (UC) demonstrated proteins primarily involved immune response, cell‐cell interactions extracellular matrix interactions. Our analyses unveil notable disparities associated biological functions among subpopulations, underscoring importance meticulously selecting resultant based intended application.

Abstract Multiple immune cells and their products in the liver together form a complex unique microenvironment, preclinical models have demonstrated importance of imbalances hepatic microenvironment inflammatory diseases immunocompromised diseases. Various immunotherapies been attempted to modulate for purpose treating Mesenchymal stem (MSCs) comprehensive plastic immunomodulatory capacity. On one hand, they tried treatment because excellent immunosuppressive capacity; other MSCs immune-enhancing properties settings can be modified into cellular carriers targeted transport enhancers by genetic modification, physical chemical loading, thus are also used such as chronic viral infections hepatocellular carcinoma. In this review, we discuss immunological basis recent strategies aforementioned Specifically, update summarize distinct mechanisms imbalance diseases, how fully exploit immunotherapeutic role with both patterns.

In recent years, the launch of clinical-grade exosomes is rising expeditiously, as they represent a new powerful approach for delivery advanced therapies and diagnostic purposes various diseases. Exosomes are membrane-bound extracellular vesicles that can act biological messengers between cells, in context health disease. comparison to several lab-based drug carriers, exosome exhibits high stability, accommodates diverse cargo loads, elicits low immunogenicity toxicity, therefore manifests tremendous perspectives development therapeutics. The efforts made spur drugging untreatable targets encouraging. Currently, T helper (Th) 17 cells considered most prominent factor establishment autoimmunity genetic disorders. Current reports have indicated importance targeting Th17 secretion its paracrine molecule, interleukin (IL)-17. However, present-day targeted approaches exhibit drawbacks, such cost production, rapid transformation, poor bioavailability, importantly, causing opportunistic infections ultimately hamper their clinical applications. To overcome this hurdle, potential use vectors seem be promising cell-targeted therapies. With standpoint, review discusses concept by providing snapshot biogenesis, summarizes current trials diseases, analyzes prospect an established carrier delineates present challenges, with emphasis on practical applications We further decode possible future scope bioengineering against catastrophe.
