Acute methicillin resistant Staphylococcus aureus (MRSA) pneumonia is one of the most frequently seen lung infection diseases with high morbidity and mortality. It urgent to explore an efficient antibacterial strategy owing increase drug resistance, virulence, pathogenicity MRSA. was found that Fe3O4 can induce ferroptosis in MRSA, but its effect inhibited by glutathione (GSH) a certain extent, while cinnamaldehyde (CA) enhance consuming GSH. As bacterial quorum sensing (QS) inhibitor, CA suppress QS system further exert antibiofilm effects. Here, we developed Fe3O4-based inducer promote interrupt QS, destroy biofilm, thus effectively treat acute MRSA pneumonia. We used sodium alginate (SA) wrap form particles, then coated surface hybrid biomimetic membrane composed erythrocyte platelet obtain targeted particles (mFe-CA). Under ultrasonic (US) stimulation, mFe-CA efficiently release CA, thereby synergically inducing death characteristics ferroptosis, including mass ROS production, lipid peroxidation, GSH depletion, respiratory chain suppression. Furthermore, + US inhibit system, remove biofilms, reduce strain virulence. In mouse model pneumonia, treatment markedly advanced survival rate mice, reduced load lungs, alleviated inflammatory damage, there no obvious toxicity. This study proposes substitute which may provide foreground for overcoming microbial resistance fighting biofilm-associated infections also provides target theoretical basis clinical

Abstract Psoriasis is an immune-mediated inflammatory skin disease, involving a complex interplay between genetic and environmental factors. Previous studies have demonstrated that factors play major role in the pathogenesis of psoriasis. However, non-genetic are also necessary to trigger onset recurrence psoriasis genetically predisposed individuals, which include infections, microbiota dysbiosis gut, dysregulated lipid metabolism, sex hormones, mental illness. can be induced by other triggers, such as trauma, unhealthy lifestyles, medications. Understanding how these triggers provides insights into pathogenesis, well better clinical administration. In this review, we summarize for update current evidence on underlying mechanism elicit disease.

Polystyrene microplastics (PS-MPs) are new types of environmental pollutant that have garnered significant attention in recent years since they were found to cause damage the human respiratory system when inhaled. The pulmonary fibrosis is one serious consequences PS-MPs inhalation. However, impact and underlying mechanisms on not clear. In this study, we studied potential lung toxicity PS-MPs-developed by long-term intranasal inhalation PS-MPs. results showed after exposing PS-MPs, lungs model mouse had different levels fibrosis. Meanwhile, resulted a markedly decrease glutathione (GSH), an increase malondialdehyde (MDA), iron overload tissue mice alveolar epithelial cells (AECs). These findings suggested occurrence PS-MP-induced ferroptosis. Inhibitor ferroptosis (Fer-1) alleviated PS-MPs-induced Mechanically, triggered cell promoted development via activating cGAS/STING signaling pathway. Inhibition with G150/H151 attenuated exposure. Together, these data provided novel mechanistic insights therapeutic paradigm.

Sodium aescinate (SA), a natural plant extract with various bioactivities, is widely used to treat oedema and inflammation in clinics. However, adverse events, including liver injury, kidney phlebitis, have been reported patients SA recent years. In this study, we BALB/c mice L02 cells evaluate the role of ferroptosis SA-induced injury. significantly increased AST, ALT, MDA Fe2+, decreased GSH levels, induced pathological changes vivo. also reduced viability LDH release, intracellular cysteine reduction, depletion, iron accumulation, ROS production, lipid peroxidation, indicating that causes ferroptosis. addition, inhibited transcriptional activity activating transcription factor 4 (ATF4) subsequently expression downstream genes xCT (solute carrier family 7a member 11, SLC7A11) Cystathionine gamma-lyase (CTH) which play vital roles biosynthesis. Interestingly, cytotoxic effects were effectively attenuated by ATF4 overexpression, while they aggravated silencing. These results revealed triggers hepatocyte inhibiting ATF4, an oxidative imbalance.

Background/Objective: Ferroptosis is an iron-dependent form of programmed cell death characterized by lipid peroxidation products (LPOs). A chemotherapeutic drug, 5–fluorouracil (5–FU), can induce epithelial mucositis and favor drug synergism with erastin in ferroptosis. Camellia tea saponin extract (TS) known to exert antioxidative properties. This study aims delineate the protective role TS mitigating 5–FU-induced ferroptosis inflammation human keratinocytes. Methods: HaCaT cells were induced 5–FU erastin, treated different doses, their viability was then determined. Levels cellular reactive oxygen species (ROS), LPOs, labile iron pool (LIP), glutathione (GSH), peroxidase 4 (GPX–4) activity, as well IL–6, IL–1β, TNF–α levels, wound healing properties assessed. Results: per se (at up 25 µg/mL) not toxic but unable restore baseline levels. The compound significantly diminished increases ROS, LIP, while restoring GSH content GPX–4 activity. Additionally, it suppressed cytokine production a concentration–dependent manner. Moreover, exerted wound-healing effects against skin injuries damage dose dependently.
